摘 要:目的探讨胰腺炎大鼠腹腔积液中硫氧还蛋白表达与腹膜功能衰竭的关联机制。方法60只SD大鼠按照随机数字表法分为2组:对照组(n=30,不做任何干预)和实验组(n=30,通过注射牛磺胆酸钠构建大鼠急性胰腺炎模型)。模型建立成功后次日,将每组大鼠分为2批,分别采用N-甲酰甲硫氨酰-亮氨酰-苯丙氨酸(fMLP)和硫乙醇酸盐腹腔注射,并于2种炎性介质注射后0.5、2、8.0 h获取免疫球蛋白和腹腔积液。应用组织化学染色法检测β-gal的表达,分析腹膜表面的转基因MCS的面积百分比,测定腹腔积液中硫氧还蛋白表达和二硫化物氧化还原酶的活性,评估腹膜MCS炎症的严重情况。结果炎症介质注射后0.5~8.0 h,实验组胰腺炎模型大鼠MCSβ-gal阳性面积均显著低于对照组,差异均有统计学意义(P<0.05)。炎症介质注射8 h后,实验组胰腺炎模型大鼠腹腔积液中二硫化物氧化还原酶和人硫氧还蛋白活性均较对照组显著降低,差异均有统计学意义(P<0.05)。炎症介质注射8 h后,实验组大鼠脂多糖、白细胞介素-1β和肿瘤坏死因子-α水平较对照组增加,人硫氧还蛋白表达显著降低,差异均有统计学意义(P<0.05)。结论胰腺炎大鼠腹腔积液中硫氧还蛋白表达和二硫化物氧化还原酶活性均显著下降,同时诱发腹膜功能衰竭,因此通过上调腹腔积液中人硫氧还蛋白的水平和二硫化物氧化还原酶活性可能有助于缓解腹膜间皮细胞的炎症。Objective To investigate the relationship between thioredoxin expression in peritoneal effusion and peritoneal failure in rats with pancreatitis.Methods Sixty were divided into two groups according to the random number table method(n=30,without any intervention)and experimental group(n=30,acute pancreatitis model was established by injection of sodium taurocholate).The next day after the model was successfully established,each group of rats was divided into two batches,and N-formylmethionyl-leucyl-phenylalanine(fMLP)and thioglycolate were injected intraperitoneally respectively.And at 0.5,2,8.0 h after the injection of the two inflammatory mediators,immunoglobulin and peritoneal effusion were obtained.Histochemical staining method was used to detect the expression ofβ-gal,analyze the area percentage of transgenic MCS on the surface of the peritoneum,determine the expression of thioredoxin and the activity of disulfide oxidoreductase in the peritoneal effusion,and evaluate the severity of peritoneal MCS inflammation.Results 0.5~8.0 h after the injection of inflammatory mediators,the positive area of MCSβ-gal in the experimental group of pancreatitis model rats was significantly lower than that of the control group,and the difference was statistically significant(P<0.05);Eight hours after the injection of inflammatory mediators,the activities of disulfide oxidoreductase and human thioredoxin in the ascites of pancreatitis model rats in the experimental group were significantly lower than those in the control group,and the differences were statistically significant(P<0.05).Eight hours after the injection of inflammatory mediators,the levels of lipopolysaccharide,interleukin-1β,tumor necrosis factor alpha in the experimental group were increased compared with the control group,and the expression of human thioredoxin was significantly decreased(P<0.05).Conclusion The expression of thioredoxin and disulfide oxidoreductase activity in the peritoneal effusion of pancreatitis rats were significantly decreased,and perit
