摘 要:目的探讨星形细胞上调基因-1(AEG-1)和M2型丙酮酸激酶(PKM2)在骨肉瘤组织和骨软骨瘤组织中表达及其临床意义。方法收集大庆市龙南医院(齐齐哈尔医学院第五附属医院)2014年3月至2020年11月资料完整的21例骨软骨瘤和71例骨肉瘤组织标本,使用免疫组织化学法检测AEG-1和PKM2的表达水平,并采用χ2检验进行统计学分析。结果骨肉瘤组织中AEG-1阳性表达率60.56%(43/71),明显高于骨软骨瘤组织AEG-1阳性表达率19.05%(4/21),两者差异有统计学意义(χ2=11.178,P<0.01),AEG-1表达水平与骨肉瘤恩内金分期(Eneeking分期)、肺转移明显相关(χ2=4.461、6.277,P<0.05)。骨肉瘤组织中PKM2阳性表达率66.20%(47/71),明显高于骨软骨瘤组织PKM2阳性表达率28.57%(6/21),两者差异有统计学意义(χ2=9.395,P<0.01),PKM2表达水平与骨肉瘤Eneeking分期、肺转移和软组织浸润明显相关(χ2=6.552、5.253、5.591,P<0.05)。结论AEG-1和PKM2在骨肉瘤组织中表达水平升高,AEG-1和PKM2可能与骨肉瘤的恶性程度、疾病的进程以及骨肉瘤侵袭转移密切相关。Objective To investigate the expression and clinical significance of astrocyte elevated gene-1(AEG-1)and M2-pyruvate kinase(PKM2)in osteosarcoma tissues and osteochondroma tissues.Methods A total of 21 cases of osteochondroma tissues and 71 cases of osteosarcoma tissues pathologically confirmed from March 2014 to November 2020 on Daqing Longnan Hospital(Qiqihar Medical School No.5 Affiliated Hospital)were collected.The expression of AEG-1 and PKM2 in osteochondroma tissues and osteosarcoma tissues was detected by immunohistochemistry.The statistical analysis of the data was performed using Chi-square test.Results The positive rate of AEG-1 expression in osteosarcoma tissues[60.56%(43/71)]was significantly higher than that in osteochondroma tissues[19.05%(4/21)](χ2=11.178,P<0.01).The expression of AEG-1 was related osteosarcoma to Eneeking stage,lung metastasis(respectively,χ2=4.461,6.277,P<0.05).The positive rate of PKM2 expression in osteosarcoma tissues[66.20%(47/71)]was significantly higher than that in osteochondroma tissues[28.57%(6/21)](χ2=9.395,P<0.01).The expression of PKM2 was related to Eneeking stage,lung metastasis and soft tissue involvement of osteosarcoma(respectively,χ2=6.552,5.253,5.591,P<0.05).Conclusion AEG-1 and PKM2 were upregulated in osteosarcoma tissues.The higher expression of AEG-1 and PKM2 was closely related to the degree of malignancy,disease progression,invasion and metastasis of osteosarcoma.
