摘  要:目的:探讨黄芩苷对结肠癌HT-29细胞移植瘤小鼠IL-6/STAT3信号转导通路的影响。方法:选取SPF级健康雄性昆明种小鼠96只,采用人结肠癌HT-29细胞悬液右前肢腋窝处皮下注射建立结肠癌移植瘤模型。将小鼠随机分为对照组、模型组、黄芩苷低剂量组、黄芩苷中剂量组、黄芩苷高剂量组及阳性对照组,各16只,均腹腔注射给药10 ml/kg、1次/d,连续14 d。检测比较各组肿瘤体积、肿瘤质量、抑瘤率,肿瘤组织细胞凋亡指数(AI),血清IL-6、IL-10水平,以及肿瘤组织IL-6、STAT3、p-STAT3、Bcl-2蛋白表达水平。结果:与模型组比较,各组小鼠抑瘤率、肿瘤组织AI较高,且黄芩苷低剂量组<黄芩苷中剂量组<黄芩苷高剂量组<阳性对照组,差异有统计学意义(均P<0.05)。与模型组比较,各组小鼠肿瘤体积和质量、血清IL-6、IL-10水平、肿瘤组织IL-6、p-STAT3、Bcl-2蛋白表达水平较低,且黄芩苷低剂量组>黄芩苷中剂量组>黄芩苷高剂量组>阳性对照组,差异有统计学意义(均P<0.05)。结论:黄芩苷能剂量依赖性的抑制结肠癌HT-29细胞移植瘤小鼠肿瘤生长,促进肿瘤细胞凋亡,其作用可能与抑制IL-6/STAT3信号转导通路活性有关。Objective:To analysis the effect of baicalin on IL-6/STAT3 signal transduction pathway in mice with carcinoma of colon HT-29 cell transplantation tumor.Methods:96 clean healthy male Kunming mice were selected,and established colon carcinoma transplanted by subcutaneous injection of human carcinoma of colon HT-29 cell suspension into the axilla of right forelimb.The mice were randomly divided into control group model group baicalin low-dose group baicalin medium-dose group baicalin high-dose group,and positive control group,16 mice in each group,all mice were intraperitoneally injected with 10 ml/kg once a day for 14 d.The tumor volume and quality,tumor suppressor rate,apoptotic index(AI)of tumor tissues,serum levels of IL-6 IL-10,and expression of IL-6,STAT3,p-STAT3,Bcl-2 protein in tumor tissues were detected and compared.Results:Compared with model group,the tumor suppressor rate and AI of tumor tissues were higher,and baicalin low-dose group<baicalin middle-dose group<baicalin high-dose group<positive control group.The differences were statistically significant(all P<0.05).Compared with the model control group,the tumor volume and quality,serum levels of IL-6,IL-10,and expression of IL-6,p-STAT3,Bcl-2 in tumor tissues were lower in each group,and baicalin low-dose group>baicalin middle-dose group>baicalin high-dose group>positive control group.The differences were statistically significant(all P<0.05).Conclusions:Baicalin can inhibit the growth of carcinoma of colon HT-29 cell transplantation tumor in dose-dependent manner,promotes apoptosis of tumor tissue,and the effect may be related to the inhibition of IL-6/STAT3 signal transduction pathway activity.
