摘  要:目的:探讨Neuregulin-1(NRG-1)对过氧化氢(H2O2)诱导的小鼠骨髓间充质干细胞(BMSCs)衰老的影响。方法:用H2O2诱导小鼠BMSCs衰老模型,将BMSCs分为对照组、H2O2组、H2O2+NRG-1组。采用CCK8检测细胞存活率,β-半乳糖苷酶染色法检测细胞衰老,流式细胞术检测细胞周期,实时荧光定量PCR(qPCR)法检测衰老相关基因P53、P21 mRNA的表达,Western blotting检测乙酰化(acetyl)-P53、P21的表达变化,划痕实验、Transwell实验检测细胞的迁移情况。结果:与对照组比较,H2O2组β-半乳糖苷酶染色阳性细胞百分比升高,细胞周期处于G0/G1期的细胞比例、P53、P21 mRNA表达、acetyl-P53、P21蛋白表达水平显著增高,而细胞迁移能力明显下降(均P<0.05)。与H2O2组比较,H2O2+NRG-1组β-半乳糖苷酶染色阳性细胞百分比、细胞周期处于G0/G1期的细胞比例、P53、P21 mRNA表达下调及acetyl-P53、P21蛋白表达水平显著降低,迁移能力明显增强(均P<0.05)。结论:NRG-1能延缓H2O2诱导的BMSCs衰老,并能促进BMSCs的增殖和迁移。Objective:To investigate the effect of Neuregulin-1(NRG-1)on attenuating the senescence of mouse bone marrow mesenchymal stem cells(BMSCs)induced by hydrogen peroxide(H2O2).Methods:The senescent BMSCs of mouse was constructed by H2O2,and BMSCs were divided into the control group,H2O2 group and H2O2+NRG-1 group.CCK8 was used to detect cell viability,β-galactosidase staining was used to detect cell senescence,and flow cytometry was used to detect cell cycle.The expression of P53 and P21genes was measured using qPCR.Western blotting was used to detect changes in the expression of acetyl-P53 and P21.The scratch test and transwell test were used to detect cell migration.Results:Compared with the control group,the percentage ofβ-galactosidase staining positive cells in the H2O2group increased,and the proportion of cells in the G0/G1 phase,the expression of P53 and P21 mRNA and the expression levels of acetyl-P53 and P21 proteins increased,but the migration ability decreased significantly(P<0.05).Compared with the H2O2 group,the percentage ofβ-galactosidase staining positive cells,the proportion of cells in the G0/G1 phase,the expression of P53 and P21 mRNA,and expression levels of acetyl-P53 and P21 proteins were decreased in the H2O2+NRG-1 group,whereas the migration ability was notably increased(P<0.05).Conclusion:NRG-1 can delay the aging of BMSCs induced by H2O2 and promote the proliferation and migration of BMSCs.
