摘  要:目的:利用单细胞质谱流式技术(CyTOF)研究HBV相关肝癌和非HBV相关肝癌的免疫微环境特征。方法:收集42例HCC患者的癌组织样本(28例HBV相关肝癌和14例非HBV相关肝癌)并制备成单细胞悬液。通过质谱流式技术用35种免疫细胞相关抗体对肝癌组织进行单细胞组学检测,并分析肿瘤浸润性免疫细胞中免疫相关标志物的表达情况。结果:CyTOF成功将HBV相关与非HBV相关肝癌的CD45+免疫细胞分成了21个细胞亚群。HBV相关肝癌组织与非HBV相关肝癌的免疫细胞组成存在明显差异,Th细胞在HBV相关HCC中占比最高,而Tc细胞在非HBV相关HCC中的占比最高。结论:成功构建HBV相关肝癌和非HBV相关肝癌的免疫微环境图谱,并揭示了HBV相关肝癌与非HBV相关肝癌的免疫微环境存在显著异质性。Objective:To study the characteristics of the immune microenvironment in HBV and non-HBV related hepatocellular carcinoma(HCC)using single-cell mass cytometry(CyTOF).Methods:A total of 42 HCC specimens(28 HBV-related and 14 non-HBV related HCC)were collected and then dissociated into single cell suspension.A total of 29 immune cell related antibodies were used to perform single-cell functional proteomics analysis and further analyze the expression of immune related markers in tumor infiltrating immune cells based on mass cytometry.Results:The workflow for CyTOF was successfully established and CD45+immune cells were divided into 21 cell clusters in HBV-related HCC.There were significant differences in the composition of immune cells between HBV and non-HBV related HCC.Moreover,Th cells were accounted for the highest proportion in HBV related HCC,while Tc cells were accounted for the highest proportion in non-HBV related HCC.Conclusion:The profile of immune microenvironment in HBV and non-HBV related HCC was successfully constructed,and the significant heterogeneity in HBV and non-HBV related HCC was innovatively revealed.
