补肾益气和血方对卵巢储备功能下降大鼠血清性激素和卵巢组织VEGF及其受体蛋白表达的影响

摘　　要：目的观察补肾益气和血方对卵巢储备功能下降大鼠血清性激素及卵巢组织血管内皮生长因子(VEGF)及其受体蛋白表达的影响。方法将48只雌性SD大鼠随机分为6组各8只。空白组给予生理盐水15 mL/kg灌胃;其他组均上午给予雷公藤多苷50 mg/kg灌胃,下午模型组给予生理盐水灌胃,补佳乐组给予补佳乐0.2 mg/kg灌胃,补肾益气和血方低、中、高剂量组分别给予4.31 g/kg、8.62 g/kg、17.25 g/kg补肾益气和血方灌胃。连续灌胃21 d后,显微镜下观察各组大鼠阴道涂片动情周期及卵巢组织形态学变化;ELLSA法测定血清促卵泡生成素(FSH)、黄体生成素(LH)、雌二醇(E2)、抗苗勒管激素(AMH)水平;免疫组织化学法测定卵巢组织中VEGF及其受体flt-1、kdr蛋白表达情况。结果与空白组比较,模型组大鼠动情周期明显延长,卵巢指数及血清E2、AMH水平均明显降低(P均<0.05),血清FSH水平和FSH/LH均明显升高(P均<0.05);与模型组比较,补肾益气和血方高剂量组大鼠卵巢指数和补肾益气和血方中、高剂量组及补佳乐组血清E2、AMH水平均明显升高(P均<0.05),补肾益气和血方中、高剂量组血清FSH水平和补肾益气和血方高剂量组FSH/LH均明显降低(P均<0.05);补肾益气和血方中、高剂量组血清FSH水平均明显低于补佳乐组(P均<0.05);补肾益气和血方高剂量组和补佳乐组血清E2水平均明显高于补肾益气和血方低、中剂量组(P均<0.05),补肾益气和血方高剂量组和补佳乐组血清E2、AMH水平比较差异均无统计学意义(P均>0.05)。与空白组比较,模型组大鼠卵巢组织中VEGF及flt-1、kdr蛋白表达积分吸光度值均明显降低(P均<0.05);与模型组比较,补肾益气和血方各剂量组和补佳乐组大鼠卵巢组织中VEGF及flt-1、kdr蛋白表达积分吸光度值均明显升高(P均<0.05),补肾益气和血方各剂量组和补佳乐组间比较差异均无统计学意义(P均>0.05)。结论补肾益气和血方Objective It is to observe the effects of Bushen Yiqi Hexue Decoction on serum sex hormones and expression of vascular endothelial growth factor(VEGF)and its receptor protein in ovarian tissue in rats with decreased ovarian reserve.Methods Forty-eight female SD rats were randomly divided into 6 groups with 8 rats in each group.The blank group was given normal saline 15 mL/kg by gavage;the other groups were given tripterygium polyglycosides 50 mg/kg by gavage in the morning,furthermore,the model group was given normal saline by gavage,the Bujiale group was given Bujiale 0.2 mg/kg by gavage,and the low,medium and high dose groups of Bushen Yiqi Hexue Decoction were respectively given Bushen Yiqi Hexue Decoction 4.31 g/kg,8.62 g/kg,17.25 g/kg by gavage in the afternoon.After continuous gavage for 21 days,the estrus cycle and the changes of ovarian tissue morphology of the rats in each group were observed under microscope;the levels of serum follicle-stimulating hormone(FSH),luteinizing hormone(LH)and estradiol(E2)and anti-Müllerian hormone(AMH)were determined by ELLSA method;the expression of VEGF and its receptor flt-1 and kdr protein in ovarian tissue were detected by immunohistochemical method.Results Compared with the blank group,the estrous cycle of rats in the model group was significantly prolonged,the ovarian index and the levels of serum E2 and AMH were significantly reduced(all P<0.05),and the levels of serum FSH and FSH/LH were significantly increased(all P<0.05);compared with the model group,the ovarian index of rats in the Bushen Yiqi Hexue Decoction high-dose group and the levels of serum E2 and AMH in the medium and high-dose Bushen Yiqi Hexue Decoction and Bujiale group were significantly increased(all P<0.05),the levels of serum FSH in the medium and high dose groups of the Bushen Yiqi Hexue Decoction and the FSH/LH of the Bushen Yiqi Hexue Decoction high-dose group were significantly reduced(all P<0.05);the levels of serum FSH of the medium and high-dose groups of Bushen Yiqi Hexue Decoction were
