摘  要:目的对驱虫斑鸠菊种子类黄酮物质进行定量分析。方法采用选择性反应/多反应监测技术(SRM/MRM),利用超高效液相色谱及MRM质谱分析,对驱虫斑鸠菊种子类黄酮物质进行定量分析,并结合KEGG数据库,对驱虫斑鸠菊种子类黄酮化合物的参与的主要分子生物学过程进行分析。结果测定驱虫斑鸠菊种子总黄酮类物质含量为0.97 g/100g,通过高效液相色谱及MRM质谱分析,共获得36种黄酮类化合物,含量前5位的化合物为:圣草酚(12650.98 ng/100g)、异槲皮苷(7293.86 ng/100 g)、木樨草素(4503.38 ng/100 g)、紫铆素(4320.83 ng/100 g)和柚皮素(3390.59 ng/100 g),而异黄酮化合物芒柄花黄素和异黄酮化合物黄豆素含量最少,仅占1.07 ng/100 g和1.81 ng/100 g。驱虫斑鸠菊种子类黄酮组分涉及的生物合成过程主要包括类黄酮生物合成、黄酮和黄酮醇生物合成、异黄酮生物合成、泛醌和其它萜类生物合成及酪氨酸代谢等。结论本研究应用SRM/MRM技术,该方法特异性强、灵敏度高、准确性高,适用于对特定复合物组分进行有针对性地、特异性地检测与分析,并可获得目标化合物的绝对定量结果,也为驱虫斑鸠菊化学成分进一步研究奠定基础。Objective To quantify analysis the flavonoid constituents in the ethanol extract of Vernonia anthelmintica(L.)Willd.Methods Selective reaction/multiple response monitoring technique(SRM/MRM)combined high performance liquid chromatograph(HPLC)and MRM mass spectrometry were used for quantitative analysis of the flavonoid constituents.Combined KEGG database,the biosynthesis process of each component was analyzed.Results The content of total flavonoids of Vernonia anthelmintica(L.)Willd.is 0.97 g/100g,and a total of 36 flavonoid compounds were quantified from the ethanol extract of Vernonia anthelmintica(L.)Willd.The top 5 compounds in the list of contents were holophenol(12650.98 ng/100g),isoquercitrin(7293.86 ng/100g),luteolin(4503.38 ng/100g),rivetin(4320.83 ng/100g)and naringin(3390.59 ng/100g),and the last 2 compounds were Formononetin(1.07 ng/100g)and Daidzin(1.81 ng/100g).The biosynthesis processes of flavonoid components in the seeds of Vernonia anthelmintica(L.)Willd include flavonoid biosynthesis,flavonoid and flavonol biosynthesis,isoflavone biosynthesis,ubiquinone and other terpene biosynthesis and tyrosine metabolism,and etc.Conclusion The method of SRM/MRM is strong specificity,high sensitivity and high accuracy,which can be detected and analyzed in a targeted and specific way,and can obtain the absolute quantitative results of the target metabolite,which also lays the fundation for further research on the chemical components of Vernonia anthelmintica(L.)Willd.
