摘 要:目的通过30天喂养试验,分析虾青素软胶囊对大鼠的毒性作用,为开发虾青素提供实验室依据。方法选用80只离乳SD大鼠,雌雄各40只,单笼喂养,自由地进食和饮水。试验设立3个剂量组:83、167、333 mg/(kg·bw)。同时设食用植物油对照组。连续30天每日1次性经口灌胃。观察大鼠每日的行为、一般表现、有无中毒表现以及是否存在死亡情况。每周称重1次并计算进食量和食物利用率。试验结束处死大鼠,采血进行血液学和生化学的各项指标测定。对大鼠进行解剖,观察大鼠内脏的改变。称取脏器重量,进行脏体比计算。摘取肝脏、肾脏、脾脏、胃脏、肠和睾丸(卵巢)等组织进行病理学的检查。结果连续灌胃30天后,雌雄大鼠未发生死亡,亦未出现明显中毒症状。大鼠体重持续性增加,生长良好。各剂量组大鼠的体重、每周进食量和食物利用率、血液生化指标、脏体比与阴性对照组相比,差异均无统计学意义(均P>0.05)。血液学指标除雄鼠低剂量组的血红蛋白略高于阴性对照组(t=3.894,P<0.05),中剂量组血红蛋白和嗜碱细胞略高于阴性对照组(t=3.640、3.078,均P<0.05)外,其余指标与阴性对照组比较,差异均无统计学意义(均P>0.05)。组织病理学结果显示未见高剂量组有中毒性的损伤改变。结论30天喂养试验后,虾青素软胶囊对大鼠无明显毒性作用。Objective To study the toxicity of astaxanthin soft capsule on rats by 30-day feeding test,provide laboratory basis for the development of astaxanthin.Methods Totally 80 mature and healthy SD rats were selected,40 males and 40 females,and were fed and drank freely in single cage.Three dose groups were set up:83,167,333 mg/(kg·bw).A control group of edible vegetable oil was set.Oral gavage was administered once a day for 30 consecutive days.The daily behavior,general manifestations,toxicity and death of the rats were observed.The animals were weighed once a week,and food intake and food utilization were calculated.The rats were killed at the end of the experiment,and the blood samples were collected to test the hematological and biochemical indicators.The rats were dissected to observe the changes of viscera.The organs were weighed to calculate the visceral ratio.Liver,kidney,spleen,stomach,intestine,testis(ovary)and other tissues were extracted for pathological examination.Results After 30 days of continuous gavage,there was no death and no obvious poisoning symptoms in both male and female rats.The rats continued to gain weight and grew well.There were no statistically significant differences in body weight,food intake per week,food utilization rate,blood biochemical indexes and visceral ratio between each dose group and the negative control group(all P>0.05).For the hematological indicators,the hemoglobin in the low dose group of male rats was slightly higher than that in the negative control group(t=3.894,P<0.05),and the hemoglobin and basophilic cells in the middle dose group were slightly higher than those in the negative control group(t=3.640,3.078,both P<0.05),and the differences in other indicators were not statistically significant between the dose groups and the negative control group(all P>0.05).Histopathological results showed no toxic injury changes were observed in the high dose group.Conclusion After 30 days of feeding,astaxanthin soft capsule has no obvious toxicity on rats.
