摘  要:目的了解阿尔茨海默病(AD)大鼠口服补阳还五汤后,其大脑海马区基质金属蛋白酶-9(MMP-9)的水平和钙蛋白酶活性的变化。方法制作AD大鼠模型,因不同的处理方式将实验设计为4组:正常组,AD组,补阳还五汤组和脑复康组。补阳还五汤组和脑复康组分别服用补阳还五汤和脑复康,正常组和AD组服用同等量的生理盐水,服用时长为4周。观察4组大鼠的行为能力的变化;免疫印迹技术分析大鼠海马区MMP-9的值;试剂盒检测大鼠海马区钙蛋白酶的活性变化。结果和正常组比较,AD组大鼠的行为能力变弱,海马区MMP-9的值升高(0.715±0.037),钙蛋白酶活性增强(2.741±0.384),差异均有统计学意义(均P<0.05);和AD组比较,补阳还五汤组和脑复康药组大鼠的行为能力提高,补阳还五汤组海马区MMP-9的值降低(0.338±0.016),钙蛋白酶活性变弱(1.821±0.149),差异均有统计学意义(均P<0.05)。结论补阳还五汤能改善AD大鼠的行为能力,这与其能减少AD大鼠海马区MMP-9的量和减弱钙蛋白酶活性密切相关。Objective To understand the changes in the levels of matrix metalloproteinase-9(MMP-9)and calpain activity in the hippocampus of Alzheimer’s disease(AD)rats after oral administration of Buyang Huanwu Decoction.Methods AD rat model was made,and the experiment was designed into 4 groups according to different treatment methods:normal group,AD group,Buyang Huanwu Decoction group and Naofukang medicine group.Buyang Huanwu Decoction group and Naofukang group were given Buyang Huanwu Decoction and Naofukang respectively for 4 weeks.The normal group and AD group were given the same amount of normal saline for 4 weeks.The change of behavior ability of four groups of rats were observed,the MMP-9 of hippocampus was analyzed by Western blot,and the changes of calpain activity in hippocampus of rats were detected by kit.Results Compared with the normal group,the behavior ability of rats in AD group became weak,the value of MMP-9 in the hippocampus increased(0.715±0.037),the activity of calpain increased(2.741±0.384),and the differences were statistically significant(all P<0.05).Compared with the AD group,the behavior ability of rats in Buyang Huanwu Decoction group and Naofukang group was improved,the Buyang Huanwu Decoction group value of MMP-9 in hippocampus was decreased(0.338±0.016),and the activity of calpain was weakened(1.821±0.149),the differences were all statistically significant(all P<0.05).Conclusion Buyang Huanwu Decoction can improve the behavior of AD rats,which is closely related to its ability to reduce the amount of MMP-9 and weaken the activity of calpain in the hippocampus of AD rats.
