摘  要:目的探讨Ras效应因子-1(RIN1)在上皮性卵巢肿瘤中的表达水平及意义。方法收集辽宁省肿瘤医院自2017年1月至2018年12月收治的150例接受手术治疗患者的新鲜卵巢组织标本。150例患者中,正常卵巢30例(A组),上皮性卵巢良性肿瘤30例(B组),上皮性卵巢交界性肿瘤30例(C组),上皮性卵巢恶性肿瘤60例(D组)。D组中,病理分期Ⅰ+Ⅱ期30例,Ⅲ+Ⅳ期30例;分化程度高-中分化30例,低分化30例。采用Western Blot法检测RIN1蛋白表达水平,聚合酶链式反应(PCR)法检测RIN1 mRNA表达水平。结果 A组、B组、C组、D组RIN1蛋白相对表达量分别为(0.096 2±0.026 2)、(0.371 0±0.028 3)、(0.564 3±0.056 7)、(0.732 0±0.098 2),两两比较,差异均有统计学意义(P<0.05);D组中,Ⅲ+Ⅳ期RIN1蛋白相对表达量高于Ⅰ+Ⅱ期[(0.813 0±0.061 5)比(0.651 0±0.047 6)],低分化RIN1蛋白相对表达量高于高-中分化[(0.785 0±0.087 9)比(0.679 0±0.078 0)],差异均有统计学意义(P<0.05)。A组、B组、C组、D组RIN1 mRNA相对表达量分别为(3.890 0±0.440 5)、(5.517 0±0.281 7)、(7.593 0±0.636 8)、(9.722 0±1.177 0),两两比较,差异均有统计学意义(P<0.05);D组中,Ⅲ+Ⅳ期RIN1 mRNA相对表达量高于Ⅰ+Ⅱ期[(10.710 0±5.248 0)比(8.733 0±0.723 7)],低分化RIN1 mRNA相对表达量高于高-中分化[(10.450 0±0.895 9)比(8.990 0±0.953 9)],差异均有统计学意义(P<0.05)。结论随着上皮性卵巢肿瘤恶性程度上升,RIN1表达明显增强,RIN1在上皮性卵巢恶性肿瘤的发生和发展过程中具有重要的促进作用。Objective To investigate the expression of Ras interactor-1(RIN1)in epithelial ovarian tumors.Methods A total of 150 fresh ovarian tissue specimens were collected from 150 patients who underwent surgical treatment in Liaoning Cancer Hospital and Institute from January 2017 to December 2018.Among the 150 patients, 30 were normal ovarian(Group A),30 were epithelial benign ovarian tumors(Group B),30 were epithelial borderline ovarian tumors(Group C),and 60 were epithelial malignant ovarian tumors(Group D).In Group D,30 patients were diagnosed with pathological stage Ⅰ+Ⅱ,30 patients were Ⅲ+Ⅳ,while, there were 30 cases with high-medium differentiation and 30 cases with low differentiation.The protein expression level of RIN1 was detected by Western Blot, and the mRNA expression level of RIN1 was detected by polymerase chain reaction(PCR).Results The protein expression level of RIN1 in groups A,B,C and D were(0.096 2±0.026 2),(0.371 0±0.028 3),(0.564 3±0.056 7)and(0.732 0±0.098 2),respectively, and the differences were statistically significant(P<0.05).In group D,the protein expression level of RIN1 at Ⅲ+Ⅳ stage was higher than that at Ⅰ+Ⅱ stage[(0.813 0±0.061 5)vs.(0.651 0±0.047 6)],the protein expression level of RIN1 in low differentiation was higher than that in high-medium differentiation[(0.785 0±0.087 9)vs.(0.679 0±0.078 0)],P<0.05.The mRNA expression level of RIN1 in groups A,B,C and D were(3.890 0±0.440 5),(5.517 0±0.281 7),(7.593 0±0.636 8)and(9.722 0±1.177 0),respectively, and the differences were statistically significant(P<0.05).In group D,the mRNA expression level of RIN1 at Ⅲ+Ⅳ stage was higher than that at Ⅰ+Ⅱ stage[(10.710 0±5.248 0)vs.(8.733 0±0.723 7)],the mRNA expression level of RIN1 in low differentiation was higher than that in high-medium differentiation[(10.450 0±0.895 9)vs.(8.990 0±0.953 9)],P<0.05.Conclusion The expression of RIN1 was enhanced with the increase of malignant degree of epithelial ovarian tumors, RIN1 played an important role in promoting
