摘 要:原花青素抗氧化程度受原花青素平均聚合度的影响很大,低聚原花青素的活性明显高于高聚原花青素的活性。从花生红衣提取物中所得到的原花青素相当一部分是以高聚体的形式存在,将高聚体通过水解,变成低聚体,对于增加人体吸收效率和植物的利用度都有较大作用。以花生红衣原花青素提取物为原料,经水溶解平衡后过滤分离,滤去低聚体,对高聚体原花青素分别使用磷酸、柠檬酸及732型阳离子树脂等为催化剂进行水解。着重研究温度和时间对水解反应的影响,水解后原花青素平均聚合度的测定采用硫酸香草醛法。研究表明,在相同条件下,732型阳离子树脂是最适宜的水解催化剂。通过单因素和正交试验得到优化的水解反应条件:原花青素甲醇溶液与阳离子树脂体积比为2.0∶1、水解温度60℃下,反应15 min的平均聚合度为2.55。The antioxidation of proanthocyanidins was greatly affected by the average degree of polymerization of proanthocyanidins,and the activity of proanthocyanidins was significantly higher than that of high procyanidins.A large part of proanthocyanidins obtained from peanut red coat extracts existed in the form of highpolymers.The hydrolysis of highpolymer into oligomer has a great effect on the absorption efficiency and the physical utilization of the plant.The proanthocyanidin extract of peanut red coat was used as raw material.The high proanthocyanidins were hydrolyzed by phosphoric acid,citric acid and 732 cationic resin separately through water dissolution equilibrium separation and filtration of oligomers.The temperature and time of hydrolysis reaction were studied.The average degree of polymerization of proanthocyanidins after hydrolysis was determined by sulfuric acid-vanillin method.The results show that 732 cationic resin is the most suitable hydrolysis catalyst under the same conditions.By single factor and orthogonal experiment,the optimum reaction conditions of hydrolysis were obtained as follows:the average degree of polymerization of 15 min was 2.55 at 60℃and at 2.0∶1 volume ratio of procyanidin methanol solution to cationic resin.
