摘 要:目的评估不同处理下的急性早幼粒白血病(acute promyelocytic leukemia, APL)细胞来源的微粒(microparticle, MP)对脐静脉内皮细胞的损伤作用。方法提取5例APL患者的骨髓单核细胞,体外单独培养48 h,或者与全反式维甲酸、三氧化二砷、柔红霉素共培养24 h,收集细胞培养液提取MP,将提取后的微粒与人脐静脉内皮细胞(HUVECs)共培养24 h,用ELISA,流式细胞术测内皮活化指标,用复钙时间法测HUVECs的促凝活性。结果与对照组相比,APL源性的微粒显著促进HUVECs的活化,与单纯的APL细胞源性的微粒相比,柔红霉素和三氧化二砷增加了MP对HUVECs的损伤作用。结论 APL源性微粒能够损伤HUVECs,增加促凝活性。Objective To evaluate the effect of microparticles(MP) derived from acute promyelocytic leukemia(APL) in different treatments on umbilical vein endothelial cells. Methods Bone marrow mononuclear cells extracted from 5 cases of APL patients were cultured 48 h alone, or with all trans retinoic acid and arsenic trioxide, or daunorubicin for 24 h, respectively cell culture fluid was collected for the extraction of MP. Human umbilical vein endothelial cells(HUVECs) were co-cultured with the particles for 24 h. ELISA and flow cytometry test were used to measure endothelial activation index, complex calcium method was performed for coagulant activity time. Results Compared with the control group, APL particles significantly promoted the activation of HUVECs. Compared with pure APL cell-derived particles, daunorubicin and arsenic trioxide increased the damage of MP on HUVECs. Conclusion APL-derived MPs can damage HUVECs and increase their procoagulant activity.
