摘　　要：目的探讨长链非编码RNA(long non-coding RNA,LncRNA)SOX21-AS1在宫颈癌中的表达及其临床诊断价值。方法收集宫颈脱落细胞学标本共43例,其中正常23例,宫颈癌20例。采用实时定量聚合酶链反应(real-time quantitative polymerase chain reaction, qRT-PCR)法检测所有标本中LncRNA SOX21-AS1的表达水平,分析LncRNA SOX21-AS1的表达与宫颈癌患者各项临床病理特征的关系及其对宫颈癌的诊断价值。结果宫颈癌组LncRNA SOX21-AS1的表达水平显著高于正常组,差异有统计学意义(P=0.000)。随患者年龄增大、肿瘤直径增加、FIGO分期增高、浸润深度增加及出现淋巴结转移,LncRNA SOX21-AS1表达量呈增高趋势,其中按淋巴结是否转移及FIGO分期分组,LncRNA SOX21-AS1表达量组间差异具有显著性(P=0.001;P=0.025);与之相反,随组织学分级增高,LncRNA SOX21-AS1表达下降,组间差异有统计学意义(P=0.008)。受试者工作特征曲线(receiver operating characteristic curve, ROC)分析LncRNA SOX21-AS1表达对宫颈癌的诊断价值结果显示,ROC曲线下面积(area under the curve, AUC)为0.911(95%CI:0.826~0.996,P=0.000),当约登指数取最大值时,LncRNA SOX21-AS1表达水平诊断宫颈癌的敏感性为70%,特异性为100%。结论 LncRNA SOX21-AS1在宫颈癌中显著高表达,具有较高的临床诊断价值,有望成为宫颈癌诊疗新的生物标志物。Objective To investigate the expression of long non-coding RNA(LncRNA) SOX21-AS1 in cervical cancer and its clinical significance. Methods A total of 43 samples of cervical liquid-based cytology were collected, of which 23 were normal samples and 20 were cervical cancer samples. The real-time quantitative polymerase chain reaction(qRT-PCR) was used to detect the expression level of LncRNA SOX21-AS1 in all specimens. In addition, The diagnostic value of LncRNA SOX21-AS1 expression in cervical cancer and the relationship between expression level and clinicopathological characteristics of cervical cancer patients were analyzed. Results The expression levels of LncRNA SOX21-AS1 in cervical cancer group were significantly higher than those in normal group, and the difference was statistically significant(P=0.000). The expression of LncRNA SOX21-AS1 increased with the increase of age, tumor diameter, FIGO stage, lymph node metastasis and depth of invasion, but only according to lymph node metastasis and FIGO stage, the expression of LncRNA SOX21-AS1 was significantly different between the two groups(P=0.001;P=0.025), on the contrary, the expression of LncRNA SOX21-AS1 decreased with the increase of histological grade, and the difference was statistically significant(P=0.008). The value of LncRNA SOX21-AS1 in diagnosis of cervical cancer was analyzed by receiver operating characteristic curve(ROC). The results showed that the area under the curve(AUC) was 0.911(95% CI: 0.826~0.996, P=0.000). When the Jordan index reached the maximum, the sensitivity of LncRNA SOX21-AS1 expression in the diagnosis of cervical cancer was 70%, and the specificity was 100%. Conclusion LncRNA SOX21-AS1 is highly expressed in cervical cancer and have high diagnostic value for cervical cancer, and is expected to become new biomarker for cervical cancer diagnosis and treatment of cervical cancer.
