摘 要:目的探讨微小核糖核酸-34(miR-34)与其靶基因生存素(survivin)在人结肠癌中的表达及意义。方法收集32例手术患者切除的新鲜组织,其中32份结肠癌组织标本(均为腺癌)作为癌症组,32份癌旁组织标本(据切缘>2 cm)作为癌旁组。采用荧光定量聚合酶链式反应(PCR)技术检测miR-34和survivin基因在癌症组和癌旁组中的转录表达水平,并进行比较。结果miR-34a和miR-34b/c基因表达在癌症组组织中均低于癌旁组,差异有统计学意义(P<0.05);在癌症组组织中miR-34b/c表达较miR-34a水平降低更显著,survivin表达增加较癌旁组增高,差异有统计学意义(P<0.05);miR-34a和miR-34b/c与survivin在结肠癌癌组织中表达水平呈显著负相关(r=-0.43、-0.51,P<0.05)。结论miR-34家族在结肠腺癌中作为抑癌基因发挥作用,可以负性调节survivin的表达,为结肠癌的临床诊治提供新的靶点和思路。Objective To discuss the expression and significance of microRNA-34(miR-34)and survivin gene in human colon cancer tissue.Methods Of the 32 fresh tissues removed by surgery,32 colon cancer tissues(all adenocarcinoma)were used as the cancer group,and 32 adjacent tissues(according to the margin>2 cm)were used as paracancerous group.The transcriptional expression levels of miR-34 and survivin genes in cancer group and paracancerous group were detected and compared by fluorescence quantitative polymerase chain reaction(PCR).Results The expression of miR-34 a and miR-34 b/c genes in the cancer group was lower than that in the adjacent group,and the difference was statistically significant(P<0.05).The expression of miR-34 b/c in the cancer group was significantly lower than that of miR-34 a,and the increase in survivin expression was higher than that in the adjacent group.All the difference was statistically significant(P<0.05).The expression levels of miR-34 a and mir-34 b/c in tissues of colon cancer group were negatively correlated with that of survivin(r=-0.43,-0.51,P<0.05).Conclusion The miR-34 family plays a role as the tumor suppressor gene in colorectal adenocarcinoma,which can negatively regulate the expression of survivin,and provide new ideas for the clinical diagnosis and treatment of colorectal adenocarcinoma.
