摘 要:目的研究人乳头瘤病毒(HPV)感染对宫颈癌(CC)组织程序性死亡受体-1(PD-1)、程序性死亡受体配体-1(PD-L1)和叉头翼状螺旋转录因子(FOXP3)蛋白表达的影响.方法选取2016年1月-2019年1月甘肃省妇幼保健院妇瘤科接受治疗的CC患者89例为研究对象,检测宫颈癌组患者中HPV-16/18感染情况,并根据感染情况分为HPV感染组61例和HPV非感染组28例.采用免疫组化法检测两组FOXP3、PD-1及PD-L1表达情况.并根据免疫组化评分将89例宫颈癌患者分为FOXP3、PD-1和PD-L1高表达组和低表达组,分析FOXP3、PD-1、PD-L1表达与临床病理特征的关系.结果61例HPV感染患者其中HPV-16感染30例,HPV-18感染31例;HPV感染组患者PD-1、PD-L1和FOXP3免疫组化评分分别为(6.85±1.26)分、(4.65±0.69)分和(3.04±0.51)分,显著高于HPV非感染组(P均<0.05);FOXP3、PD-1、PD-L1蛋白表达与肿瘤直径无相关性,FOXP3、PD-1、PD-L1蛋白表达与患者不同FIGO分期、组织学分级、是否存在淋巴结转移有关(P<0.05).结论HPV感染组宫颈癌组织中FOXP3、PD-1及PD-L1蛋白表达高于HPV非感染组,且与患者FIGO分期、分级、淋巴转移关系密切.OBJECTIVE To observe the influence of human papillomavirus(HPV)infection on expression of programmed death 1(PD-1),programmed death ligand 1(PD-L1)and forkhead winged helix transcription factor(FOXP3)proteins in cervical cancer(CC)tissues.METHODS Totally 89 CC patients who were treated in gynecological oncology department of Gansu Maternal and Child Health Hospital from Jan 2016 to Jan 2019 were recruited as the study subjects,the prevalence of HPV-16/18 infection among the CC patients was detected,and the CC patients were divided into the HPV infection group with 61 cases and the no HPV infection group with 28 cases.The expression levels of FOXP3,PD-1 and PD-L1 were detected by using immunohistochemistry,89 CC patients were divided into the FOXP3,PD-1 and PD-L1 high expression groups and low expression groups according to immunohistochemical score,and the correlation between the expression of FOXP3,PD-1 and PD-L1 and the clinical pathological features was observed.RESULTS Of 61 patients with HPV infection,30 had HPV-16 infection,and 31 had HPV-18 infection.The immunohistochemical scores of PD-1,PD-L1 and FOXP3 of the HPV infection group were respectively(6.85±1.26)points,(4.65±0.69)points and(3.04±0.51)points,significantly higher than those of the no HPV infection group(all P<0.05).There was no correlation between the expression of FOXP3,PD-1 and PD-L1 proteins and the diameter of tumor;the expression of FOXP3,PD-1 and PD-L1 proteins was associated with the FIGO stage,histological grade and lymph node metastasis(P<0.05).CONCLUSION The expression levels of FOXP3,PD-1 and PD-L1 proteins in CC tissues are higher in the HPV infection group than in the no HPV infection group,which are closely associated with the FIGO stage,histological grade and lymph node metastasis.
