摘  要:目的探讨微小RNA(miR)-561调控E2F6蛋白表达对神经干细胞增殖和分化的影响及其可能的作用机制。方法采用实时荧光定量PCR技术(qPCR)检测E2F6蛋白在胎鼠中的表达水平;分化病毒感染后的神经干细胞,采用荧光免疫染色和qPCR检测E2F6蛋白过表达对神经干细胞分化的影响。结果E2F6蛋白在胎鼠大脑各个区域和其他器官中的表达量明显不同,其中大脑区域,E2F6蛋白在海马和小脑两个区域的表达量相对较高,而在中脑表达量最低;E2F6蛋白可促进神经干细胞向少突胶质细胞分化。结论通过对miR-561调控E2F6蛋白表达对神经干细胞增殖和分化的影响及其作用机制的研究,可能有效改善肌萎缩侧索硬化症、多发性硬化症等相关疾病水平。Objective To investigate the effect of microRNA(miRNA,miR)-561 on the differentiation and proliferation of NSCs regulated by E2 F6 protein expression and its possible mechanism.Methods Detecting the E2 F6 protein expression in mice by qPCR.At the same time,the virus-infected neural stem cells were differentiated and the effect of E2 F6 overexpression on the differentiation of neural stem cells was detected by fluorescence immunostaining and qPCR,respectively.Results The expression of E2 F6 protein in various regions of the mouse brain and other organs was significant different.In the brain region,the expression of E2 F6 protein was relatively higher in the hippocampus and cerebellum and the midbrain expression was lower.The E2 F6 protein in the nerve stem cells had a certain role in promoting oligodendrocyte differentiation.Conclusion The miR-561 has effect on the differentiation and proliferation of NSCs by regulating E2 F6 protein expression,which may effectively improve the level of oligodendrocyte related diseases such as myotrophic gurat sclerosis,sababaraha sclerosis.
