摘  要:目的分析前列腺癌组织中人吻素1(KISS-1)和G蛋白偶联受体54(GPR54)的表达水平,并探讨二者与前列腺癌患者临床病理特征及预后的关系。方法选取郑州大学附属郑州中心医院2016年4月至2018年5月收治的30例前列腺癌患者为前列腺癌组,另选取同期前列腺增生患者20例作为对照组。采用免疫组化法检测两组中KISS-1和GPR54蛋白表达情况;采用定量反转录聚合酶链式反应(qRT-PCR)检测两组KISS-1和GPR54 m RNA的表达水平;分析KISS-1和GPR54表达与前列腺癌组织临床病理特征及患者预后的关系。结果免疫组化结果显示,前列腺癌组中KISS-1阳性表达率低于对照组(16.67%vs.80.00%)(P<0.05),GPR54阳性表达率与对照组差异无统计学意义(P>0.05)。qRT-PCR结果显示,前列腺癌组中KISS-1 mRNA表达量低于对照组[(0.62±0.06)vs.(0.88±0.09)](P<0.05),GPR54 mRNA表达量与对照组差异无统计学意义(P>0.05)。KISS-1和GPR54阳性表达与年龄、淋巴结是否转移无关,KISS-1和GPR54在不同TNM分期下的阳性表达差异有统计学意义(P<0.05),GPR54在不同Gleason评分下的阳性表达差异有统计学意义(P<0.05)。KISS-1阳性表达者1年生存率高于阴性表达者(100.00%vs.80.00%),GPR54阳性表达者1年生存率低于阴性表达者(80.77%vs.100.00%),差异均有统计学意义(P<0.05)。结论KISS-1和GPR54在前列腺组织中异常表达可能与前列腺癌的发生发展及患者预后有关,有望成为预测前列腺癌的潜在生物指标。Objective To analyze the expressions of people kiss element 1(KISS-1)and G protein coupled receptor 54(GPR54)in prostate cancer,and explore the relationship between KISS-1,GPR54 and the clinicopathological characteristics and prognosis of prostate cancer.Methods A total of 30 patients with prostate cancer admitted to Zhengzhou Central Hospital Affiliated to Zhengzhou University,from April 2016 to May 2018 were selected as the prostate cancer group,and 20 patients with prostatic hyperplasia who came to our hospital in the same period were selected as the control group.The expressions of KISS-1 and GPR54 were detected by immunohistochemistry;quantitative reverse transcription polymerase chain reaction(qRT-PCR)was used to detect the expressions of KISS-1 and GPR54 m RNA in the two groups,and the relationship between the expressions of KISS-1 and GPR54 and the clinicopathological characteristics and prognosis of prostate cancer were analyzed.Results Immunohistochemistry showed that,the positive expression rate of KISS-1 in prostate cancer group was lower than that in control group(16.67%vs.80.00%)(P<0.05),while there was no statistically significant difference between prostate cancer group and control group(P>0.05);qRT-PCR results showed that the expression of KISS-1 mRNA in prostate cancer was lower than that in control group[(0.62±0.06)vs.(0.88±0.09)](P<0.05),while there was no statistically significant difference in GPR54 mRNA expression(P>0.05).The positive expressions of KISS-1 and GPR54 were not related to age and lymph node metastasis,there was significant difference in the positive expressions of KISS-1 and GPR54 among different TNM stages(P<0.05),and the positive expression of GPR54 in different Gleason scores was statistically significant(P<0.05).The survival rate of KISS-1 positive patients was higher than that of negative patients(100.00%vs.80.00%)(P<0.05),and the survival rate of GPR54 positive patients was lower than that of negative patients(80.77%vs.100.00%)(P<0.05).Conclusion Abnormal expression of
