摘  要:目的探究维生素检测培养基质量和配方对维生素B12含量测定的影响。方法比较不同配方菌株活化培养基、种子液制备肉汤培养基、不同品牌测定培养基对维生素B12结果测定的影响。利用莱士曼氏乳杆菌对维生素B12的特异性和灵敏性,定量测定质控样品1849A中维生素B12的含量,并通过质量控制措施对测定结果进行分析评价。结果检测过程中使用BD品牌的维生素B12琼脂和维生素B12接种肉汤培养基、B1品牌的维生素B12测定培养基得到的拟合曲线线性更好,质控样品维生素B12检测结果组内差异也更小。结论培养基质量控制对维生素B12检测的准确度和稳定性极为重要。Objective To explore the influence of the quality and formula of the vitamin detection media on the determination of vitamin B12 content.Methods The effects of different formulations of strain activation media,seed liquid preparation broth media,and different brands of determination media on the determination of vitamin B12 results were compared.Using the specificity and sensitivity of Lactobacillus leishmani to vitamin B12,the content of vitamin B12 in the quality control sample 1849 A was quantitatively determined,and the determination results were analyzed and evaluated through quality control measures.Results In the detection process,the fitting curve obtained by using BD brand vitamin B12 agar,vitamin B12 inoculation broth media and B1 brand vitamin B12 determination media was more linear,and the differences in the results of the vitamin B12 test of the quality control samples were also smaller within the group.Conclusion Media quality control is extremely important to the accuracy and stability of vitamin B12 detection.
