摘  要:目的探究CXC趋化因子受体4(CXCR4)及其配体趋化因子基质细胞衍生因子-1(SDF-1)在成釉细胞瘤(AB)中的表达及意义。方法实时定量PCR、Western blotting及免疫组化方法检测2007年至2012年中国医科大学口腔医院病理科30例AB,10例正常口腔黏膜(NOM)冻存组织样本以及80例AB及10例口腔鳞状细胞癌(OSCC)存档蜡块中CXCR4及SDF-1 mRNA及蛋白表达情况,并进行统计分析。结果实时定量PCR结果显示,CXCR4及SDF-1在AB中相对表达量均高于NOM,差异有统计学意义(均P<0.05);Western blotting结果显示,AB中CXCR4及SDF-1蛋白表达明显高于NOM,差异有统计学意义(P<0.05);免疫组化结果显示,AB中CXCR4表达呈明显阳性(70/80),主要在细胞质中表达,NOM表达呈阴性;SDF-1表达呈明显阳性(68/80),主要定位于细胞质,NOM表达呈阴性。结论AB中CXCR4及其配体表达明显高于正常口腔黏膜,可能参与介导AB侵袭复发等异常生物学行为。Objective To investigate the expression and significance of CXC chemokine receptor 4(CXCR4)and its ligand stromal cell derived factor-1(SDF-1)in ameloblastoma(AB).Methods The mRNA and protein expressions of CXCR4 and SDF-1 were detected by QRT PCR,Western blotting,and immunohistochemistry in 10 cases of normal oral mucosa(NOM),80 cases of AB,and 10 cases of OSCC.Results QRT PCR results showed that the relative expression of CXCR4 and SDF-1 in AB was higher than that in NOM(P<0.05).Western blotting results showed that the expression of CXCR4 and SDF-1 in AB was significantly higher than that in NOM(P<0.05).Immunohistochemistry results showed that CXCR4 expression in AB was significantly positive(70/80),mainly in the cytoplasm,and that expression in NOM was negative.SDF-1 expression was significantly positive(68/80),also mainly in the cytoplasm,while NOM expression was negative.Conclusion The expression of CXCR4 and its ligand in AB is significantly higher than that in NOM.This may lead to AB invasion and other abnormal biological behaviors.
