摘  要:目的观察含铜L605钴基合金材料对血管内皮化的影响。方法将体外培养的人脐静脉内皮细胞(HUVEC)随机分为L605-2.0 Cu组、L605-2.5 Cu组、L605-3.5 Cu组及L605组,分别给予含铜量为2.0wt%、2.5wt%及3.5wt%的L605钴基合金及单纯L605钴基合金干预;采用细胞计数试剂盒(CCK-8)检测细胞增殖活力[光密度(OD)值)],以Transwell小室实验检测细胞迁移能力(穿膜细胞数),以小管生成实验检测血管形成能力(小管生成总长度及节点数)。结果以不同含铜量L605钴基合金干预细胞48 h后,L605-3.5 Cu组OD值高于L605组(P<0.05);L605-2.5 Cu组、L605-2.0 Cu组与L605组OD值差异均无统计学意义(P均>0.05)。不同含铜量L605钴基合金浸提液与细胞共培养24 h后,各组细胞形态均良好;穿膜细胞数L605-3.5 Cu组高于L605组和L605-2.0 Cu组(P均<0.01),L605-2.0 Cu组与L605组差异无统计学意义(P>0.05);L605-3.5 Cu组小管生成总长度及节点数均高于其他3组(P均<0.01),L605-2.5 Cu组、L605-2.0 Cu组与L605组差异均无统计学意义(P均>0.05)。结论含铜L605钴基合金材料促进血管内皮化,以含铜量为3.5wt%的L605-3.5 Cu效果最显著。Objective To observe the impacts of copper-containing cobalt-based alloy materials on endothelialization.Methods Human umbilical vein endothelial cells(HUVEC)cultured in vitro were randomly divided into L605-2.0 Cu group,L605-2.5 Cu group,L605-3.5 Cu group and L605 group,and L605 cobalt base alloy with copper content of 2.0wt%,2.5wt%,3.5wt%and L605 cobalt base alloy alone were given to different groups,respectively.The cell counting kit-8(CCK-8)was used to evaluate the cell proliferation activity(optical density[OD]value),while the Transwell method was used to investigate the cell migration(the number of penetrating cells),and the tubule formation assay was used to examine the blood vessel formation ability(total tube length and branching points).Results The OD value of L605-3.5 Cu group was higher than that of L605 group after 48 h of L605 cobalt-based alloy with different copper content(P<0.05),while no statistical difference of OD value was found among L605-2.5 Cu group,L605-2.0 Cu group and L605 group(all P>0.05).After co-culture with L605 cobalt alloy extracts with different copper contents for 24 h,the cell morphology of all groups were good,the number of penetrating cells in L605-3.5 Cu group was more than that in L605 group and L605-2.0 Cu group(both P<0.01),but there was no statistical difference between L605-2.0 Cu group and L605 group(P>0.05).The total length of tubule formation and the number of nodes in L605-3.5 Cu group were both larger than those in the other 3 groups(all P<0.01),while there was no significance difference among L605-2.5 Cu group,L605-2.0 Cu group and L605 group(all P>0.05).Conclusion Copper-containing L605 cobalt-based alloy material promoted vascular endothelialization,and L605-3.5 Cu with copper content of 3.5wt%had the most significant effect.
