摘 要:目的探讨枸杞多糖对大鼠心肌梗死后交感神经重构的影响。方法将大鼠随机分为心肌梗死模型组、枸杞多糖组、假手术组,每组20只。对左冠状动脉前降支穿线结扎制备心肌梗死模型。程序刺激各组大鼠诱发室性心律失常进行易感性测定,免疫组织化学方法检测各组大鼠心肌梗死周边区组织TH和GAP43的阳性神经纤维密度,Western Blot检测各组大鼠心肌梗死周边区NGF蛋白的表达情况。结果模型组室性心率失常诱发率明显高于假手术组,枸杞多糖组室性心率失常诱发率较模型组有明显下降(P<0.05);模型组阳性神经纤维密度较假手术组有明显增多,枸杞多糖组阳性神经纤维密度较模型组有所下降(P<0.05);模型组大鼠梗死周围区NGF蛋白表达显著高于假手术组,枸杞多糖组大鼠梗死周围区NGF蛋白表达低于心梗模型组(P<0.05)。结论枸杞多糖可改善大鼠心肌梗死后交感神经重构,降低室性心律失常发生率。Objective To investigate the effect of Lycium barbarum polysaccharide on sympathetic nerve remodeling after myocardial infarction in rats.Methods The rats were randomly divided into themodel group,Lycium barbarum polysaccharide group and sham operation group,20 rats in each group.The model of myocardial infarction was prepared by threading and ligating the anterior descending branch of the left coronary artery.Susceptibility determination was done by procedural stimulation of rats in each group to induce ventricular arrhythmia.Immunohistochemical methods were used to detect the positive nerve fibers density of TH and GAP43 around myocardial infarction tissues.Western Blot was used to detect the NGF protein expression of around myocardial infarctiontissue.Results The induction rate of ventricular arrhythmia in themodel group was significantly higher than that in the sham operation group,and the induction rate in the Lycium barbarum polysaccharide group was significantly lower than that in the model group(P<0.05).The density of positive nerve fibers in the model group was significantly higher than that in the sham operation group,and the density of positive nerve fibers in the Lycium barbarum polysaccharide group was lower than that in the model group(P<0.05).The expression of NGF protein around myocardial infarction tissues in the model group was significantly higher than that of the sham operation group,and the expression of NGF protein in the Lycium barbarum polysaccharide group was lower than that of the model group(P<0.05).Conclusion Lycium barbarum polysaccharide can improve sympathetic nerve remodeling after myocardial infarction in rats and reduce the incidence of ventricular arrhythmia.
