摘 要:目的研究腹外侧眶额皮质5-HT2A受体表达水平对福尔马林诱导的口面部炎性痛发生的影响,揭示组)、5-HT2A受体敲减组(KD组);腹外侧眶额皮质注射腺病毒建立小鼠5-HT2A受体敲减模型;4周后采用Western印迹法验证受体敲减效果;然后用微量注射器将30μl 1%福尔马林溶液(或生理盐水)注射到小鼠触须垫,记录45 min小鼠前、后爪摩擦触须垫时间。结果 Western印迹结果表明,KD组5-HT2A受体表达水平明显低于WT组和NC组,表明5-HT2A受体敲减成功。行为学结果表明在早期时相,福尔马林组小鼠摩擦触须垫的总时间无统计学差异,NC组之间无统计学差异。结论腹外侧眶额皮质5-HT2A受体可能在小鼠福尔马林炎性痛模型中通过影响晚期时相摩擦行为参与口面部疼痛调节。Objective To explore the effects of 5-HT2Areceptors on formalin-induced orofacial inflammatory pain in the ventrolateral orbitofrontal cortex(VLO). Methods C57BL male mice aged 6-8 weeks were randomly divided into three groups(WT for wild-type,NC for empty vector group,and KD for 5-HT2Areceptor knockdown group). Adenovirus was injected into the ventrolateral orbital cortex to establish a knockdown model of 5-HT2Areceptors. After 4 weeks,Western blotting was used to evaluate the expression of 5-HT2Areceptor before 30 μl of 1% formalin solution or normal saline was injected into the mouse’s whisker pad with a micro syringe. The amount of time mice spent rubbing the whisker pads with fore or hind paws within 45 minutes was recorded. Results Western blotting results showed the expression of 5-HT2Areceptor in the KD group was significantly lower than that of the WT and NC group,indicating that 5-HT2Areceptor was knocked down in the ventrolateral orbital cortex. Behavioral experimental results showed that in the early phase,there was no statistically significant differences in the rubbing time of the whisker pads between KD,WT,and NC groups,but the time was longer than in the saline group. In the late phase,the rubbing time in the KD group was significantly longer than in the NC and WT groups,but there were no significant differences between the WT group and the NC group after injection of formalin solution. Conclusion The 5-HT2Areceptor in the ventrolateral orbital cortex may be involved in the regulation of orofacial pain in the model of formalin-induced inflammatory pain by affecting the rubbing behavior in the late phase.
