摘  要:目的分析柑橘橘络提取物中的类黄酮成分并探究其抗低氧作用。方法采用高效液相色谱(HPLC)法分析和定量柑橘橘络提取物中常见类黄酮的种类和含量。60只雄性BALB/c小鼠,随机分为空白对照组、柑橘橘络提取物组、3种类黄酮组、9种类黄酮组,灌胃15 d后,进行小鼠常压密闭低氧实验,观察各组溶液对小鼠抗低氧能力的影响。结果成分分析显示,柑橘橘络提取物中类黄酮以橙皮苷、槲皮素、柚皮芸香苷为主,其他含量较高的类黄酮为:橙皮素、柚皮素,新橙皮苷、柚皮苷、坎二菲酮、杨梅黄酮含量较低,川陈皮素、橘皮素、玉米黄酮、芹菜配基未检出。常压密闭低氧实验结果显示,与空白对照组比较,15 mg/kg的柑橘橘络提取物组、3种类黄酮组、9种类黄酮组分别使小鼠常压密闭环境下的生存时间延长15.3%、16.7%和21.4%,且均有统计学意义。结论柑橘橘络提取物延长小鼠常压密闭环境下的生存时间可能主要与提取物中富含类黄酮,特别是橙皮苷、槲皮素、柚皮芸香苷有关,且类黄酮之间可能存在协同作用。Objective To analyze the components of citrus tangerine pith extract and to explore their anti-hypoxia effect. Methods High-performance liquid chromatography(HPLC)was used to analyze and determine the content of flavonoids in citrus tangerine pith extract. Sixty BALB/c mice were randomly divided into four groups:blank control group,citrus tangerine pith extract group,3-flavonoids group,and 9-flavonoids group. After 15 days of intragastric administration,a normal pressure airtight hypoxia model of mice was established,and the effects of solution from each group on anti-hypoxia ability of mice were observed. Results The results of components analysis showed that hesperidin,quercetin and narirutin were the main flavonoids in the citrus tangerine pith extract,and that other flavonoids with higher content were hesperetin and naringenin. The contents of neohesperidin,naringin,kaempferol and myricetin were relatively low,but no nobiletin,tangeretin,luteolin or apigenin were detected. Compared with the blank control group,citrus tangerine pith extract,3-flavonoids and 9-flavonoids significantly prolonged the survival time of mice exposed to normal pressure airtight hypoxia by 15.3%,16.7% and 21.4%,respectively. Conclusion The effect of citrus tangerine pith extract on prolonging the survival time of mice exposed to normal pressure airtight hypoxia may be mainly related to the high content of flavonoids,especially hesperidin,quercetin and naringin,and there may be synergistic effect among flavonoids. This study provides reference for determining the effective anti-hypoxia components in citrus tangerine pith extract.
