摘 要:通过单因素实验考察提取次数、超声波处理时间、甲醇体积浓度、超声波功率和料液比对致命鹅膏(Amanita exitialis)α-鹅膏毒肽提取得率的影响,获得关键因素及其最佳条件,再采用Box-Benhnken响应面法优化α-鹅膏毒肽超声波提取工艺。研究结果表明:致命鹅膏α-鹅膏毒肽最佳提取工艺为提取2次、超声波处理15 min、甲醇体积浓度35%、超声波功率300 W、料液比1∶20(g∶mL),α-鹅膏毒肽提取得率为3.33 mg·g^(-1)干重,与预测值3.30 mg·g^(-1)干重无显著差异(P>0.05),说明获得的最佳提取工艺稳定、可行。Using single-factor experiments,ultrasonic treatment time,volumetric methanol concentration,ultrasonic power,material to liquid ratio,and number of repeats of the extraction were studied for their effects on Amanita exitialisα-amanitin yield so as to obtain an optimal condition for each key extraction factor.Then the Box-Benhnken response surface method was used to optimize the ultrasonic extraction process.The results showed that the optimal extraction process for A.exitialisα-amanitin was as follows:ultrasonic treatment time of 15 min,35%(V∶V)methanol,300 W ultrasonic power,material-to-liquid ratio of 1∶20(g∶mL),and 2 times of repeats of the extraction.Using the optimal process,the peptide yield was 3.33 mg·g^(-1)dry weight,which was close to the predicted value of 3.30 mg·g^(-1)dry weight(P>0.05).This optimal extraction process is stable and feasible.
