摘　　要：目的:建立同时测定乌药汤配方颗粒(乌药、当归、木香、香附和甘草)中4种指标性成分含量的方法。方法:采用HPLC法,将乌药汤配方颗粒按组方比例称取并混匀,用50%甲醇超声提取,过0.22μm微孔滤膜,得提取液。分析采用Agilent ZORBAX Extend-C18(4.6 mm×250 mm,5μm)色谱柱;流动相:乙腈-0.5%磷酸,梯度洗脱;柱温:30℃;检测波长:280nm;流速:1.0mL/min。结果:去甲异波尔定、阿魏酸、甘草苷和甘草酸单铵盐分别在1.484.0 mg/L(r=1.0000)、0.41712.500 mg/L(r=1.0000)、3.9260.0 mg/L(r=1.0000)和8.4280.0 mg/L(r=0.9999)范围内呈良好的线性关系,平均加样回收率分别为100.05%、100.25%、101.41%和100.19%,RSD分别为2.51%、2.93%、2.57%和2.30%。结论:该方法简便、快速、准确、重复性好,可用于乌药汤配方颗粒的质量控制。Objective:To establish a method to simultaneously determine the contents of 4 index components in Wuyao Tang formula granules.Methods:Using the HPLC method,the Wuyao Tang formula granules were weighed and mixed according to the proportion of the prescription,extracted with 50%methanol ultrasonically,and passed through a 0.22μm microporous membrane to obtain the extract.The analysis adopts Agilent ZORBAX Extend-C18(4.6 mm×250 mm,5μm)chromatographic column.The mobile phase was acetonitrile-0.5%phosphoric acid.The flow rate was 1.0 mL/min in a gradient elution manner,column temperature was 30℃,and the detection wavelength was set at 280 nm.Results:Norisoboldine,ferulic acid,liquiritin and ammonium glycyrrhizinate showed a good linear relationship in the range of 1.4-84.0 mg/L(r=1.0000),0.417-12.500 mg/L(r=1.0000),3.9-260.0 mg/L(r=1.0000)and 8.4-280.0 mg/L(r=0.9999),respectively.The average sample recovery rates were 100.05%,100.25%,101.41%,and 100.19%,and the RSD were 2.51%,2.93%,2.57%,and 2.30%,respectively.Conclusion:The method is simple,rapid,accurate and reproducible,and can be used for the quality control of Wuyao Tang formula granules.
