摘  要:以龙胆草为发酵底物,龙胆苦苷为评价指标,采用正交实验优化龙胆酵素的发酵工艺,用高效液相色谱(HPLC)分析龙胆酵素中龙胆苦苷含量,并评估其生化功效。结果表明,最优龙胆酵素发酵工艺为菌株组合酿酒酵母加植物乳杆菌1∶1、接种量10%、发酵温度30℃、发酵时间72 h。在此优化工艺条件下,发酵液龙胆苦苷含量达到(1823±21)μg/mL,其透明质酸酶抑制率、DPPH、·OH和O^(-)_(2)·自由基的清除能力分别达到89.2%、77.5%、70.2%和74.1%,表明该工艺的发酵液具有很好的抗敏和自由基清除功效。Taking gentian as fermentation substrate and gentiopicroside as index,the fermentation process of gentian ferment was optimized by single factor and orthogonal experiments,and the gentiopicroside content in gentian ferment was analyzed by HPLC,meanwhile its biochemical efficacy was evaluated.The results showed that the optimized fermentation process of the gentian ferment were as follows:compound strain Saccharomyces cerevisiae and Lactobacillus plantarum ratio 1∶1,inoculum size 10%,fermentation temperature 30℃and fermentation time 48 h.Under the optimal conditions,the gentiopicroside content of the gentian ferment was(1823±25)μg/mL.The fermentation liquor had good anti-allergic and free radicals clearance.The hyaluronic acid enzyme inhibition rate,DPPH,·OH and O^(-)_(2) radicals clearance rates were 89.2%,77.5%,70.2%and 74.1%,respectively.
