摘 要:目的:研究宫颈鳞癌组织中SLC5A8基因甲基化状态,以及SLC5A8基因甲基化与高危型HPV的相关性。方法:随机选取2017年1月至2020年6月于湖北医药学院附属东风医院妇科住院患者共77例,术前均行HPV核酸分型检测。收集宫颈鳞状细胞癌石蜡标本36例(其中31例合并高危型HPV阳性、5例高危型HPV阴性)。收集慢性宫颈炎石蜡标本41例(其中31例合并高危型HPV阳性、10例高危型HPV阴性)。采用甲基化特异性PCR检测SLC5A8基因的甲基化状态。结果:宫颈鳞癌组中共检测到30例(83.3%)发生SLC5A8基因甲基化,宫颈炎组中检测到6例(14.6%)发生SLC5A8基因甲基化,宫颈鳞癌组的基因甲基化阳性率显著高于宫颈炎组(P<0.05)。在宫颈鳞癌组中,31例高危型HPV阳性标本中检测到28例(90.3%)发生SLC5A8基因甲基化,5例高危型HPV阴性标本中检测到2例(40%)发生SLC5A8基因甲基化,高危型HPV阳性的标本SLC5A8基因甲基化显著高于高危型HPV阴性的标本(P<0.05)。在宫颈炎组中,31例高危型HPV阳性标本中有3例(9.6%)发生SLC5A8甲基化,10例高危型HPV阴性标本中有3例(30%)发生SLC5A8甲基化,二者的SLC5A8基因甲基化差异无统计学意义(P>0.05)。结论:SLC5A8基因甲基化水平升高与宫颈鳞癌有相关性。宫颈鳞癌组织中,SLC5A8基因甲基化水平升高与高危型HPV有相关性。Objective To study the methylation status of SLC5A8 gene in cervical squamous cell carcinoma and the correla⁃tion between SLC5A8 gene methylation and high-risk HPV.Methods A total of 77 inpatients in the Department of Gynecol⁃ogy,Dongfeng Hospital of Hubei University of Medicine from January 2017 to June 2020 were randomly selected,and HPV nucleic acid typing was performed before surgery.Collect 36 paraffin specimens of cervical squamous cell carcinoma(inclu⁃ding 31 cases with high-risk HPV positive and 5 with high-risk HPV negative).Collect 41 paraffin specimens of chronic cervicitis(including 31 cases with high-risk HPV positive and 10 high-risk HPV negative).Methylation-specific PCR was used to detect the methylation status of the SLC5A8 gene.Results SLC5A8 gene methylation was detected in 30 cases(83.3%)in the cervical squamous cell carcinoma group,and SLC5A8 gene methylation was detected in 6 cases(14.6%)in the cervical squamous cell carcinoma group.The positive rate of gene methylation in cervical squamous cell carcinoma group was significantly higher than that in cervicitis group(P<0.05).In cervical squamous cell carcinoma group,SLC5A8 gene methylation was detected in 28(90.3%)of 31 high-risk HPV positive samples and 2(40%)of 5 high-risk HPV negative samples.The methylation of SLC5A8 gene in high-risk HPV positive samples was significantly higher than that in high-risk HPV negative samples(P<0.05).In cervicitis group,3(9.6%)of 31 high-risk HPV positive samples had SLC5A8 methylation,and 3(30%)of 10 high-risk HPV negative samples had SLC5A8 methylation.There was no signifi⁃cant difference in SLC5A8 gene methylation between the two groups(P>0.05).Conclusion Increased levels of SLC5A8 gene methylation are correlated with cervical squamous cell carcinoma.In cervical squamous cell carcinoma,the increased levels of SLC5A8 gene methylation are associated with high-risk HPV.
