摘  要:甲基营养型芽孢杆菌(Bacillus methylotrophicus)Hg18是一种新型生防类细菌,能产生多种抗菌活性物质。通过50%(NH4)2SO4饱和溶液沉淀透析除盐,从甲基营养型芽孢杆菌Hg18中分离到抗菌粗蛋白,经凝胶过滤层析、离子交换层析、变性聚丙烯酰胺凝胶电泳纯化得到抗菌蛋白,并对抗菌蛋白的性质进行了探究。结果表明,该抗菌蛋白对尖孢镰刀菌有很强的抑菌活性,且抑菌活性具有较好的热稳定性、酸碱稳定性、紫外线稳定性和光稳定性,可以作为潜在的生防资源进行开发与应用。Bacillus methylotrophicus Hg18 is a new type of biocontrol bacterium,which can produce a variety of antibacterial substances.In this paper,we isolated the crude antibacterial protein from Bacillus methylotrophicus Hg18 by precipitation,dialysis,desalination with 50%(NH4)2SO4 saturated solution.Moreover,we purified the crude antibacterial protein by gel filtration chromatography,ion exchange chromatography,and denaturing polyacrylamide gel electrophoresis.Furthermore,we explored the properties of antibacterial protein.The results show that the antibacterial protein has strong antibacterial activity against Fusarium oxysporum,and the antibacterial activity has good thermal stability,acid-base stability,ultraviolet stability,and light stability,which can be used as a potential biocontrol resource for development and application.
