摘 要:以70%乙醇为提取溶剂,采用回流法提取洋甘菊总黄酮,经D-101大孔吸附树脂纯化、干燥,得到洋甘菊总黄酮,含量为56%;采用液相色谱飞行时间质谱联用仪(LC-TOF-MS/MS)对纯化的洋甘菊总黄酮进行成分分析;以4-MUO为底物,测定了洋甘菊总黄酮对胰脂肪酶的抑制作用。成分分析表明,从洋甘菊总黄酮中共鉴定出38个黄酮类化合物,为质量控制提供了依据;抑制实验结果表明,洋甘菊总黄酮对胰脂肪酶具有较强的抑制作用,IC50值为(1.09±0.08)μg·mL^(-1),是安全有效的胰脂肪酶抑制剂,有望成为预防和治疗肥胖的替代药物。We extracted total flavonoids from Matricaria chamomilla L.by a reflux method with 70%ethanol as an extraction solvent,and obtained the total flavonoids from Matricaria chamomilla L.with content of 56%after purification via D-101 macroporous adsorption resin and drying.Moreover,we analyzed the total flavonoids from Matricaria chamomilla L.by LC-TOF-MS/MS.Furthermore,we determined the inhibitory effect of total flavonoids from Matricaria chamomilla L.on pancreatic lipase with 4-MUO as a substrate.The results of component analysis show that 38 kinds of flavonoids are identified from the total flavonoids from Matricaria chamomilla L.,which provide the basis for quality control.The results of inhibition experiment show that the total flavonoids from Matricaria chamomilla L.have a strong inhibitory effect on pancreatic lipase,and the IC50 value is(1.09±0.08)μg·mL^(-1),indicating the total flavonoids from Matricaria chamomilla L.is a kind of safe and effective pancreatic lipase inhibitor,which is expected to become an alternative drug for the prevention and treatment of obesity.
