摘  要:目的研究非编码长链RNA H19(Long noncoding RNA H19,LncRNA H19)对结肠癌细胞增殖、凋亡的影响以及作用机制。方法以结肠癌HT-29细胞作为研究对象,将Smart silencer NC、Smart silencer H19、siR-NC、siR-STAT1、siR-NC+Smart silencer H19、siR-STAT1+Smart silencer H19分别转染至结肠癌细胞,利用四甲基偶氮唑盐比色法(methyl thiazolyl tetrazolium,MTT)检测结肠癌细胞增殖情况;Real time PCR检测LncRNA H19、STAT1以及凋亡相关蛋白表达。结果LncRNA H19敲低可抑制结肠癌细胞增殖和Bcl-2、STAT1表达,而上调Bax表达,并且这种由LncRNA H19敲低所引发的生长抑制和促进凋亡的作用可通过STAT1敲低所增强。结论LncRNA H19可能通过STAT1促进结肠癌细胞增殖,抑制细胞凋亡。Objective To study the effects of Long non-coding RNA H19(LncRNA H19)on the proliferation and apoptosis of colon cancer cells and the mechanism of action.Methods Colon cancer HT-29 cells were transfected with Smart silencer NC,Smart silencer H19,siR-NC,siR-STAT1,siR-NC+Smart silencer H19,iR-STAT1+Smart silencer H19,respectively.Methyl Thiazolyl Tetrazolium(MTT)was used to detect the proliferation of colon cancer cells.Real time PCRwas used to detect LncRNA H19,STAT1,and apoptosis-related proteins.Results LncRNA H19 knockdown inhibited the proliferation of colon cancer cells and the expression of Bcl-2 and STAT1,while up-regulated the expression of Bax,and the effects of growth inhibition and apoptosis induced by the knockdown of LncRNA H19 could be enhanced by STAT1 knockdown.Conclusion LncRNA H19 may promote the proliferation and inhibit the apoptosis of colon cancer cells through STAT1.
