摘  要:目的在体内外探讨小檗胺对肝癌细胞SMMC-7721的增殖、迁移的影响及其可能的作用机制。方法在体外,用MTT试验评价小檗胺对SMMC-7721增殖的影响。划痕试验评价小檗胺对SMMC-7721细胞迁移的影响,在体内,用移植肿瘤模型评价小檗胺对肿瘤的增长及转移的影响。用western blot试验检测小檗胺对OTUB1、p53、Bcl-2、Bax以及caspase3表达的影响。结果MTT实验表明小檗胺以浓度依赖和时间依赖的形式抑制SMMC-7721细胞增殖;划痕试验表明小檗胺能够显著抑制SMMC-7721细胞的迁移。体内实验结果表明,小檗胺能够使肿瘤体积明显缩小,并且能够使肝脏表面的结节数明显减少,Western blot试验结果提示小檗胺能够明显下调OTUB1和Bcl-2/Bax的表达,上调cleaved-caspase-3/caspase-3和p53的表达。结论小檗胺能够在体内外SMMC-7721细胞增殖与侵袭转移,其作用机制可能与调节OTUB1的表达有关。Objective To investigate the inhibitory effects of berberine(BBM)on smMC-7721 cell proliferation and its possible action mechanism in vivo and in vitro.Methods MTT assay was used to evaluate the effects of BBM on the proliferation of SMMC-7721 in vitro,and wound healing assay was used to evaluate the effects of BBM on the metastasis of SMMC-7721 cells.And the transplantation tumor models were established in vivo to evaluate the effects of BBM on the growth and metastasis of the tumors.Moreover Western Blot was used to detect the effects of BBM on the expression levels of OTUB1,p53,Bcl-2,Bax and Caspase-3.Results The MTT assay showed that BBM inhibited the proliferation of SMMC-7721 cells in a concentration-dependent and time-dependent pattern.The wound healing assay showed that BBM significantly inhibited the metastasis of SMMC-7721 cells.The in vivo results showed that the tumor volume was significantly decreased,and the numbers of nodules on the surface of liver were significantly decreased.The Wstern Blot showed that the expression levels of OTUB1 and Bcl-2/Bax were significantly down-regulated,however,the expression levels of cleaved caspase-3/caspase-3 and p53 were significantly up-regulated.Conclusion BBM can inhibit the proliferation and metastasis of SMMC-7721 cells both in vitro and in vivo,and its action mechanism may be related with regulating the expression of OTUB1.
