摘　　要：目的探讨长链非编码RNA(long non-coding RNA,lncRNA)SOX21-AS1及SOX21在宫颈癌中的表达及其临床意义。方法收集宫颈脱落细胞学标本共43例,其中正常23例,宫颈癌20例。采用实时定量聚合酶链反应(real-time quantitative polymerase chain reaction,qRT-PCR)检测所有标本中SOX21-AS1及SOX21的表达水平,分析SOX21-AS1及SOX21的表达水平与宫颈癌患者临床病理特征的关系及其对宫颈癌的诊断价值。结果宫颈癌组SOX21-AS1和SOX21的表达水平均显著高于正常对照组,差异有统计学意义(P=0.000)。相关分析显示,SOX21-AS1与SOX21的表达量呈显著正相关(P=0.000)。SOX21-AS1与SOX21的lncRNA表达量均随年龄增大、FIGO分期增高、出现淋巴结转移及浸润深度增加而增高,其中SOX21-AS1表达量与淋巴结转移及FIGO分期增高关系最密切(P=0.001;P=0.025);SOX21表达量与FIGO分期增高关系密切(P=0.027)。与之相反,SOX21-AS1随组织学分级增高表达下降,组间差异有统计学意义(P=0.008)。受试者工作特征曲线(receiver operating characteristic curve,ROC)分析SOX21-AS1及SOX21表达在宫颈癌中诊断价值,结果显示:二者曲线下面积(area under the curve,AUC)分别为0.911(95%CI:0.826~0.996)和0.946(95%CI:0.879~1.000)(P=0.000);敏感性分别为70%和95%,特异性分别为100%和87%。结论SOX21-AS1及SOX21在宫颈癌中异常高表达,对宫颈癌具有一定的诊断价值,有望成为宫颈癌诊疗新的生物标志物。Objective To investigate the expression and clinical significance of long non-coding RNA(lncRNA)SOX21-AS1 and SOX21 in cervical cancer.Methods A total of 43 samples of cervical liquid-based cytology were collected,of which 23 were normal samples and 20 were cervical cancer samples.The real-time quantitative polymerase chain reaction(qRT-PCR)was used to detect the lncRNA expression level of SOX21-AS1 and SOX21 in all specimens.In addition,The diagnostic value of SOX21-AS1 and SOX21expression in cervical cancer and the relationship between expression level and clinicopathological characteristics of cervical cancer patients were analyzed.Results The expression levels of lncRNA SOX21-AS1 and SOX21 in cervical cancer group were significantly higher than those in normal group(P=0.000).Correlation analysis showed that there was a significant positive correlation between the expression of SOX21-AS1 and SOX21(P=0.000).The expression of SOX21-AS1 and SOX21 increased with the increase of age,FIGO stage,lymph node metastasis and depth of invasion,but only according to lymph node metastasis and FIGO stage,the expression of SOX21-AS1 was significantly different between the two groups(P=0.001;P=0.025);The expression of SOX21 increased significantly with the increase of FIGO stage(P=0.027).On the contrary,the expression of SOX21-AS1 decreased significantly with the increase of histological grade(P=0.008).The value of SOX21-AS1 and SOX21 in the diagnosis of cervical cancer was analyzed by receiver operating characteristic curve(ROC).The results showed that the area under the curve(AUC)was 0.911(95%CI:0.826~0.996)and 0.946(95%CI:0.879~1.000)(P=0.000).When the Jordan index reached the maximum,the sensitivity of expression of SOX21-AS1 and SOX21 in the diagnosis of cervical cancer was 70%and 95%,respectively,and the specificity was 100%and 87%,respectively.Conclusions SOX21-AS1 and SOX21 are highly expressed in cervical cancer and have high diagnostic value for cervical cancer,and are expected to become new biomarkers for cervical c
