摘  要:目的:探讨妊娠大鼠慢性酒精暴露对仔鼠海马发育的的影响。方法:12只妊娠期SD大鼠随机分为对照组(control)和酒精暴露组(EtOH),利用酒精灌胃的方法制备妊娠大鼠慢性酒精暴露模型,根据体重和身长评估仔鼠的发育情况,利用商品化试剂盒检测新生大鼠海马反应性活性氧(ROS)和丙二醛(MDA)的含量,利用real time RT-PCR检测海马组织中NADPH氧化酶1(NOX-1)和诱导型一氧化氮合酶(iNOS)mRNA的表达,利用Western Blot检测海马组织中凋亡相关分子cleaved caspase-3和Bcl-2的表达。结果:酒精暴露组妊娠大鼠生下的仔鼠体重和身长均小于对照组,仔鼠海马组织中ROS和MDA增加,NOX-1和iNOS mRNA的表达增加,cleaved caspase-3蛋白表达上调而Bcl-2蛋白下调。结论:妊娠大鼠慢性酒精暴露可对仔鼠海马造成损伤,其作用机制与酒精诱发氧化应激反应有关。Objective:To investigate the effects of chronic alcohol exposure on offspring hippocampus development in pregnant rats.Methods:Twelve gestational SD rats random points in the control group(control)and alcohol exposure group(EtOH).The model of chronic alcohol exposure in pregnant rats was prepared by using alcohol intragastric method.The development of the pups was assessed based on body weight and length.The levels of reactive oxygen species(ROS)and malondialdehyde(MDA)in hippocampus of newborn rats were detected by commercial kits.The mRNA expression of NADPH oxidase-1(NOX-1)and inducible nitric oxide synthase(iNOS)in hippocampus were detected by real time RT-PCR.The proteins expression of cleaved caspase-3 and Bcl-2 in hippocampus were detected by Western Blot.Results:The body weight and body length of neonatal rats in Et OH group were lower than those in the control group.Alcohol exposure during pregnancy resulted in increased ROS and MDA,increased mRNA expression of NOX-1 and iNOS,up-regulated expression of cleaved caspase-3 and down-regulated expression of Bcl-2 in the hippocampus of neonatal rats.Conclusion:Chronic alcohol exposure in pregnant rats can damage the hippocampus of neonatal rats,the mechanism of which is related to alcohol-induced oxidative stress.
