摘 要:采用分析型HPLC累积进样富集馏分的方法分离纯化干姜姜油树脂的主要化学成分,将得到的化合物用TOF/MS质谱进行鉴定,从姜油树脂中分离得到5个化合物,分别为6-gingerol;8-gingerol;diacetoxy-6-gingerdiol;6-shogaol和10-gingerol。利用MTT法对单体化合物及干姜提取物进行体外抗肿瘤活性筛选。MTT试验结果表明,6-gingerol,6-shogaol及干姜提取物对肝癌细胞均有细胞毒性,其中6-shogaol对肝癌细胞有细胞毒性且呈浓度依赖性,对SMMC-7721细胞毒性最为明显,20μM 6-shogaol作用24 h约抑制50%SMMC-7721细胞增殖。The main chemical components of ginger oleoresin were separated and purified by the method of analytical HPLC,and the compounds were identified by TOF/MS mass spectrometry.Five compounds were isolated from ginger oleoresin and identified as 6-gingerol,8-gingerol,diacetoxy-6-gingerol,6-shogaol and 10-gingerol.MTT assay was used to screen the antitumor activity of the monomers and the extract of ginger in vitro.The results showed that 6-ginger,6-shogaol and dried ginger extracts had cytotoxicity to hepatoma cells.Among them,6-shogaol had cytotoxicity on hepatoma cells in a concentration dependent manner,and the most obvious toxicity was SMMC-7721 cells.20μm 6-shogaol inhibited the proliferation of SMMC-7721 cells by 50%for 24 h.
