摘  要:目的气相色谱-质谱联用法(GC-MS)鉴定分析冠心膏中挥发性成分,并比较不同提取方法所得成分的差异。方法GC-MS条件:Thermo TG-5MS石英毛细管柱(0.25 mm×30 m,0.25μm);初始温度60℃(1 min),10℃/min升至100℃,保持30 min,7℃/min升温速率升至150℃,保持20 min,2℃/min的升温速率升至190℃,保持3 min,再以20℃/min的升温速率升至230℃,保持5 min;载气为氦气;柱流量1.0 ml/min;分流比1:1;进样口温度250℃;EI电离源70 eV;离子源温度300℃;扫描范围m/z 50~500。结果冠心膏挥发油提取法所得样品中共分离出34个峰,鉴定出19个成分,占总挥发性成分相对含量的93.4%;超声提取法所得样品中共分离出27个峰,鉴定出16个成分,占总挥发性成分相对含量的29.2%。结论两种测定方法所得冠心膏样品中挥发性成分基本一致,但所鉴定出的挥发性成分占总挥发性成分相对含量差异较大,表明使用挥发油提取法可建立更全面的冠心膏挥发性成分GC-MS信息,为冠心膏气相色谱指纹图谱的研究及其质量控制提供了依据。Objective To identify and analyze the volatile components in Guanxingao Patch by GC-MS and compare the difference of the components obtained by different extraction methods.Methods The GC-MS conditions were set as follows:A Thermo TG-5MS quartz capillary column(0.25 mm×30 m,0.25μm);the initial column temperature was 60℃and was kept for 1 min,then was raised to 100℃at a rate of 10℃/min and kept for 30 min;raised to 150℃at a rate of 7℃/min and kept for 20 min;raised to 190℃at a rate of 2℃/min and kept for 3 min;raised to 230℃at a rate of 20℃/min and kept for 5 min;helium was used as the carrier gas;the flow rate was 1.0 ml/min and the split ratio was 1:1;the inlet temperature was 250℃;the EI ionization energy was 70 eV;the ion source temperature was 300℃and the scan range was m/z 50-500.Results 34 compounds were separated from the samples obtained by extraction of volatile oil from Guanxingao Patch.Among them,19 compounds were identified,accounting for 93.4%of the relative content of the total volatile components.27 compounds were separated from the samples obtained by the ultrasonic extraction method.Among them,16 compounds were identified,accounting for 29.2%of the relative content of total volatile ingredients.Conclusion The volatile components of Guanxingao Patch obtained by the two methods are basically identical,but the relative contents of the identified compounds against total volatile components are significantly different,indicating that the volatile oil extraction can establish a more comprehensive GC-MS information of volatile components for the research of GC fingerprint and quality control of Guanxingao Patch.
