摘  要:目的:探讨miRNA-122-5p对人滑膜肉瘤细胞株SW982增殖及放疗敏感性的影响。方法:采用脂质体法分别转染miR-122-5p mimic和inhibitor至SW982细胞。流式细胞术检测细胞周期及凋亡率,MTT法检测细胞活力。通过γ-射线干预细胞,分析其对放疗的敏感性。结果:与癌旁组织相比,滑膜肉瘤miR-122-5p明显低表达,而且在转移者及低分化型中水平最低,而甲基化CpG结合蛋白2(MeCP2)的表达与之呈显著负相关(均P<0.05)。与对照组相比,miR-122-5p mimic可上调miR-122-5p水平,下调MeCP2蛋白表达,抑制SW982细胞增殖,并增加SW982细胞对射线的放疗敏感性(均P<0.05);而过表达MeCP2却能阻断miR-122-5p的作用,增加细胞对放疗的抵抗(均P<0.05)。结论:miR-122-5p在滑膜肉瘤中低表达,miR-122-5p/MeCP2轴参与调控肿瘤细胞增殖以及肿瘤细胞对γ-射线的放疗敏感性。Objective:To investigate the effect of miRNA-122-5p on the proliferation and radiotherapy sensitivity of human synovial sarcoma cell line SW982.Methods:SW982 cells were transfected with miR-122-5p mimic and inhibitor by lipofection.Flow cytometry was used to detect cell cycle and apoptosis.MTT method was used to detect cell activity.The radiotherapy sensitivity of SW982 was analyzed by γ-ray intervention.Results:The expression of miR-122-5p was significantly lower in synovial sarcoma tissue compared to the para cancer tissues,especially the lowest in metastatic and poorly differentiated types(all P<0.05).The expression of methyl-CpG-binding protein 2(MeCP2)showed a significant negative correlation with the level of miR-122-5p(P<0.05).miR-122-5p mimic significantly increased the expression of miR-122-5p in SW982(P<0.05),which remarkably decreased the proliferation rate of SW982 cells,and down-regulated MeCP2 protein expression,while increased the sensitivity of SW982 cells toγ-ray radiation(all P<0.05).However,overexpression of MeCP2 could block the effect of miR-122-5p and increased the radiation resistance(all P<0.05).Conclusion:miR-122-5p is decreased in synovial sarcoma,and the miR-122-5p/MeCP2 axis is involved in regulating the cell proliferation and the radiotherapy sensitivity of human synovial sarcoma cell.
