摘  要:目的:运用生物信息学方法筛选卵巢癌顺铂耐药差异表达基因,并对其进行相关生物学分析以获得卵巢癌耐药相关分子标志物。方法:从GEO数据库中寻找出3个卵巢癌顺铂耐药相关芯片数据集,使用GEO2R在线分析工具筛选差异表达基因,并作韦恩图获取交集;通过DAVID进行GO功能富集分析和KEGG信号通路富集分析;对显著差异表达基因构建蛋白质-蛋白质相互作用(PPI)网络图,并获取枢纽基因(Hub基因);利用UALCAN在线分析Hub基因在卵巢癌中生存曲线。结果:筛选出3个数据集的差异共表达基因67个。富集分析表明这些基因在结构分子活性、细胞对高氧反应、细胞基质粘附、成骨细胞分化、细胞外泌体、局灶性黏连、细胞外基质受体相互作用等方面具有重要功能。通过分析获得10个Hub基因:ITGB2、VCL、ITGA2、ITGB4、GBP1、COL6A1、MT2A、UST、VCAN、DSEL。生存曲线分析表明,VCAN基因表达的升高使得卵巢癌患者总体生存率显著降低(P<0.05)。结论:癌基因VCAN可作为卵巢癌顺铂耐药分子标志物,为卵巢癌顺铂耐药的诊断、治疗靶点选择及预后判断提供参考。Objective:To investigate differentially expressed genes(DEGs)with cisplatin resistance in ovarian cancer by bioinformatics methods,and to obtain molecular markers related to ovarian cancer resistance by biological analysis.Methods:Three microarray datasets of cisplatin resistance in ovarian cancer were downloaded from Gene Expression Omnibus(GEO)database.GEO2R was used to identify DEGs.The DEGs in these three datasets were screened by Venn diagram.GO function enrichment analysis and KEGG signal pathway analysis were performed using DAVID.The protein-protein interaction(PPI)network of significant DEGs was constructed,in order to get hub genes.The survival curve in ovarian cancer of hub genes were constructed using UALCAN.Results:A total of 67 significant DEGs were selected from these three microarray datasets.GO function enrichment analysis and KEGG signal pathway analysis showed that these genes have important function in structural molecular activity,cell response to hyperoxia,cell matrix adhesion,osteoblast differentiation,exosomes,focal adhesions,extracellular matrix receptor interaction,etc.There were 10 Hub genes were obtained by analysis,including ITGB2,VCL,ITGA2,ITGB4,GBP1,COL6A1,MT2A,UST,VCAN,DSEL.Survival curve analysis suggested that the increased expression of VCAN gene significantly reduced the overall survival rate of patients with ovarian cancer(P<0.05).Conclusion:VCAN can be used as a molecular marker for cisplatin resistance in ovarian cancer,providing candidate targets for diagnosis,treatment and prognostic prediction of ovarian cancer with cisplatin resistance.
