半夏泻心汤促进胃肿瘤细胞凋亡的分子机制研究

摘　　要：目的观察半夏泻心汤对胃肿瘤SGC-7901细胞的作用机制,并确定其是否可以抑制胃肿瘤细胞增殖,促进胃肿瘤细胞凋亡。方法以对数生长期的胃肿瘤SGC-7901细胞株作为研究对象,分别加入经低剂量20.25 ml/(kg·d)、中剂量40.50 ml/(kg·d)、高剂量81.00 ml/(kg·d)半夏泻心汤灌胃的大鼠血清,对照组加入经生理盐水灌胃的大鼠血清进行培养。CCK-8实验测定细胞增殖抑制率,流式细胞仪检测细胞凋亡情况和免疫印迹实验探讨凋亡作用机制。结果在CCK-8检测的结果中,低、中、高剂量组的半夏泻心汤血清处理24 h、48 h、72 h和96 h对胃肿瘤SGC-7901细胞增殖抑制率均上升,且增殖抑制率随着半夏泻心汤剂量的加大而升高(P<0.05);流式细胞检测结果显示,经过不同剂量组的半夏泻心汤血清处理48 h后,均提高了胃肿瘤SGC-7901细胞凋亡率,且凋亡率随着半夏泻心汤剂量的加大而升高(P<0.05);经半夏泻心汤的血清处理后,胃肿瘤SGC-7901细胞中B细胞淋巴瘤2(Bcl-2)的表达水平降低,而Bcl-2相关X蛋白(Bax)的表达水平增加,Bcl-2/Bax呈现出剂量依赖的方式下降(P<0.05)。结论半夏泻心汤可以抑制胃肿瘤细胞的增殖,促进凋亡。其可能的凋亡机制为通过靶向线粒体引导,发挥抗癌作用。Objective To observe the mechanism of Banxia Xiexin Decoction on gastric cancer SGC-7901 cells,and to determine whether it can inhibit the proliferation of gastric cancer cells and promote the apoptosis of gastric cancer cells.MethodsThe gastric cancer SGC-7901 cell line at the log-growth stage was used as the research object.Rat serum with low-dose 20.25 ml/(kg·d),the medium-dose 40.50 ml/(kg·d),and the high-dose 81.00 ml/(kg·d)of Banxia Xiexin Decoction were added.The control group was cultured with rat serum fed with saline.The CCK-8 assay was used to determine the inhibition rate of cell proliferation.The flow cytometry was used to detect apoptosis,and western blotting was used to investigate the mechanism of apoptosis.Results In the CCK-8 test results,the low,medium,and high-dose Banxia Xiexin Decoction serum treatment for 24 h,48 h,72 h and 96 hall increased the proliferation inhibition rate of gastric cancer SGC-7901 cells.And the proliferation inhibition rate increased with the increase of the dose of Banxia Xiexin Decoction increased(P<0.05).The results of flow cytometry showed that after 48 hours of serum treatment of Banxia Xiexin Decoction in different dose groups.The apoptosis of gastric cancer SGC-7901 cells was increased.The apoptosis rate increased with the increase of the dose of Banxia Xiexin Decoction(P<0.05).After the serum treatment of Banxia Xiexin Decoction,The expression level of B-cell lymphoma 2(Bcl-2),Bcl-2 related X protein(Bax)were increased,and the expression level of Bcl-2/Bax decreased in a dose-dependent manner(P<0.05).Conclusion Banxiaxiexin Decoction can inhibit the proliferation and promote the apoptosis of gastric cancer cells.The possible mechanism of apoptosis is to play an anticancer role by targeting mitochondrial guidance.
