摘 要:目的探讨高脂饮食对大鼠海马形态学改变及突触相关蛋白Synapsin-1(Syn-1)表达的影响。方法将18只雄性SD大鼠随机分为对照组(8只)及模型组(10只)。对照组、模型组大鼠分别给予标准饲料、高脂饲料连续饲养4周。采用化学发光法检测血清总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)的水平,ELISA法检测血清瘦素水平。使用HE染色法及镀银染色法观察海马形态学的改变,实时荧光定量PCR(qRT-PCR)及免疫组织化学技术观察海马Syn-1表达变化。结果与对照组相比,模型组大鼠血清TC、LDL及HDL浓度升高,瘦素水平降低,差异有统计学意义(P<0.01)。与对照组比较:HE染色显示,模型组大鼠海马组织的CA1、CA3及DG区神经元细胞形状不规则且分布较为松散,细胞核固缩深染;镀银染色结果显示CA1、CA3、DG脑区的神经元细胞深染加重。qRT-PCR法和免疫组织化学结果均显示,模型组中大鼠海马Syn-1的表达较对照组降低,差异有统计学意义(P<0.05或P<0.01)。结论高脂饮食可诱导大鼠高血脂症、海马神经元损伤及海马部位突触结合蛋白Syn-1表达下降。Objective To investigate the effect of high-fat diet on hippocampal morphological changes and synapse-related protein Synapsin-1(Syn-1)expression in rats.Methods 18 male SD rats were randomly divided into a control group and a model group,with 8 rats in the control group and 10 rats in the model group.Rats in the model group were given high-fat diet lasting for 4 weeks,and rats in the control group were given standard control diet.The changes of blood lipid in the rats of the two groups were detected,the morphological changes of the hippocampal tissues were observed by hematoxylin-eosin(HE)staining and silver staining,and the mRNA and protein expression of Syn-1 in the hippocampus was detected via real-time fluorescent quantitative PCR(qRT-PCR)and immunohistochemistry technique,respectively.Results Compared with those in the control group,the serum levels of total cholesterol(TC),low density lipoprotein cholesterol(LDL-C),and high density lipoprotein(HDL)in the model group significantly increased,and the level of leptin significantly reduced(P<0.01).Results of HE staining showed that the neurons in the DG,CA3 and CA1 regions of model rats were irregular in shape,loosely arranged,and hyperchromatic in nuclei.Consistently,the neurons in DG,CA3 and CA1 regions of the model rats were also hyperchromatic via silver staining.Compared with that in the hippocampus of control rats,the mRNA and protein expression of Syn-1 in the model group was significantly lower.Conclusion High-fat diet can induce hyperlipidemia,hippocampal neuron damage,and decrease expression of synaptic binding protein Syn-1 in the hippocampus of rats.
