摘 要:目的探讨环状RNA-100269(circRNA-100269)在宫颈癌中的表;达及对细胞生物学特征的影响。方法采集72例宫颈癌患者宫颈癌组织及其癌旁组织标本,检测组织中circRNA-100269表达情况,分析不同病理特征宫颈癌患者癌组织中circRNA-100269相对表达量。circRNA-100269模拟片段转染至宫颈癌细胞,分析circRNA-100269对宫颈癌细胞增殖、侵袭等生物学行为的影响。结果宫颈癌组织中circRNA-100269相对表达量明显低于癌旁组织(P <0.05)。分化程度为低分化的宫颈癌患者癌组织中circRNA-100269相对表达量明显低于分化程度为中分化、高分化的,HPV感染、有淋巴结转移、组织浸润深度>1/2患者癌组织中circRNA-100269相对表达量明显低于无淋巴结转移、组织浸润深度≤1/2(P <0.05)。转染组宫颈癌细胞增殖率明显低于对照组(P <0.05)。转染组穿膜细胞数明显小于对照组(P <0.05)。结论 circRNA-100269在宫颈癌低表达,其表达水平与宫颈癌病理特征有关,circRNA-100269对宫颈癌细胞的恶性生物学行为有一定抑制作用。Objective To investigate the expression of circular RNA-100269(circRNA-100269) in cervical cancer and its influence on cell biological characteristics.Methods Cervical cancer tissues and adjacent tissues of 72 cervical cancer patients were collected to detect the expression of circRNA-100269.The relative expression levels of circRNA-100269 in cancer tissues of patients with different pathological characteristics were analyzed.Effect of circRNA-100269 on biological behaviors of cervical cancer cells was analyzed.Results The relative expression level of circRNA-100269 was significantly lower in cervical cancer tissues than in adjacent tissues(P < 0.05).In cervical cancer patients, the relative expression level of circRNA-100269 were significantly lower in patients with low differentiation degree, HPV infection, lymph node metastasis and tissue infiltration depth > 1/2 than those with medium differentiation and high differentiation, without lymph node metastasis and with tissue infiltration depth ≤ 1/2(P < 0.05).The proliferation rate of cervical cancer cells was significantly lower in the transfection group than in the control group(P < 0.05), and number of transmembrane cells was significantly smaller than the control group(P < 0.05).Conclusions CircRNA-100269 is expression at a low level in cervical cancer, and its expression level is related to pathological characteristics of cervical cancer.In addition, circRNA-100269 has inhibitory effect on malignant biological behaviors of cervical cancer cells.
