摘 要:目的探究LncRNA MALAT1靶向miR124-3p/STAT3对宫颈癌细胞增殖和迁移的影响及其分子机制。方法采用RT-PCR检测Ect1/E6E7、Hela、SiHa和C33a细胞中LncRNA MALAT1的mRNA表达水平;构建慢病毒稳定细胞系,采用双荧光素酶报告基因分析LncRNA MALAT1和miR-124-3p-OE的靶向关系;采用MTT法检测细胞的增殖活力;采用Transwell分析细胞的侵袭情况;采用Western blot检测细胞中STAT3的蛋白表达。结果与Ect1/E6E7细胞比较,Hela、SiHa和C33a细胞中LncRNA MALAT1的mRNA相对表达水平明显上升(P <0.05)。双荧光素酶报告基因检测显示LncRNA MALAT1靶向作用miR-124-3p并下调其表达水平(P <0.05)。LncRNA MALAT1敲降或miR-124-3p过表达均可显著抑制宫颈癌细胞的增殖和侵袭,并下调STAT3的蛋白表达(P<0.05)。结论 LncRNA MALAT1通过下调miR-124-3p/STAT3分子轴促进宫颈癌细胞的增殖和迁移,有望为宫颈癌的早期诊断和临床治疗提供新的思路。Objective To investigate the effect of LncRNA MALAT1 targeting miR124-3p/STAT3 on the proliferation and invasion of cervical cancer cells and its possible molecular mechanism.Methods The mRNA expression of LncRNA MALAT1 in Ect1/E6 E7, Hela, SiHa and C33 a cells were detected by RT-PCR.The lentivirus cell lines were constructed, and the dual luciferase reporter genes were used to analyze the targeting relationship between LncRNA MALAT1 and miR-124-3p.MTT and Transwell were used to detect the cell proliferation and migration.The expression of STAT3 was analyzed by western blot.Results Compared with Ect1/E6 E7 cells, the relative expression of LncRNA MALAT1 mRNA in Hela, SiHa and C33 a cells increased significantly(P <0.05).Double luciferase reporter gene showed that LncRNA MALAT1 targeted miR-124-3p and down-regulated its expression level(P <0.05).LncRNA MALAT1 knockdown or miR-124-3p overexpression can significantly inhibit the proliferation and invasion of cervical cancer cells, and down-regulate the protein expression of STAT3(P < 0.05).Conclusion Lnc RNA MALAT1 promotes the proliferation and migration of cervical cancer cells by down-regulating the miR-124-3p/STAT3 molecular axis, which is expected to provide new ideas for the early diagnosis and clinical treatment of cervical cancer.
