健脾补肾方对化学损伤模型小鼠造血系统的影响

摘  要:目的研究健脾补肾方对化学损伤模型小鼠活性氧(ROS)及粒细胞巨噬细胞集落形成能力的影响,探讨其对化学造血系统氧化损伤的防护作用。方法取60只小鼠,随机选择12只作为正常组,另48只采用环磷酰胺腹腔注射方法建立化学损伤模型,造模成功后随机分为模型组、健脾补肾方小剂量组(100%浓度)、健脾补肾方大剂量组(200%浓度)、鲨肝醇组各12只。各药物组分别给予不同剂量健脾补肾方或鲨酐醇灌胃,正常组和模型组予以等量生理盐水灌胃。于灌胃第9天处死各组小鼠,收集外周血、骨髓。检测外周血白细胞计数、红细胞计数、血小板计数和血红蛋白水平,流式细胞仪检测骨髓中造血干/祖细胞中ROS水平,体外单层琼脂半固体培养法检测粒细胞巨噬细胞集落形成单位(CFU-GM)。结果与正常组比较,模型组小鼠外周血白细胞计数、红细胞计数、血小板计数和血红蛋白水平均明显降低(P均<0.05),骨髓细胞内ROS水平明显升高(P<0.05),骨髓细胞CFU-GM明显减少(P<0.05)。与模型组比较,健脾补肾方小、大剂量组外周血白细胞计数、红细胞计数、血小板计数和血红蛋白水平均明显升高(P均<0.05),ROS水平明显降低(P均<0.05),骨髓细胞CFU-GM明显增加(P均<0.05),其中健脾补肾方小剂量组各指标改善情况均明显优于健脾补肾方大剂量组和鲨肝醇组(P均<0.05)。结论健脾补肾方可通过降低ROS表达减轻氧化损伤,抑制造血干/祖细胞的凋亡,促进外周血白细胞、红细胞、血红蛋白、血小板的恢复来发挥对化学造血损伤的保护作用。Objective It is to investigate the effect of Jianpi Bushen Decoction(JBD)on the expression of reactive oxygen species(ROS)and granulocyte macrophage colony(CFU-GM)in chemical damage model mice,and to explore its protective effect on chemical oxidative damage and hematopoietic system damage.Methods Sixty mice were selected,in which 12 mice were randomly selected as the normal group,the other 48 mice were injected intraperitoneally with cyclophosphamide to establish chemical damage models.After successful modeling,they were randomly divided into model group,low dose JBD group(100%concentration),high dose JBD group(200%concentration),batyl alcohol(BA)group,each group had 12 mice.Each treatment group was given different doses of Jianpi Bushen Decoction or squalane alcohol by gavage,and the normal group and model group were given the same amount of normal saline gavage.On the 9th day of intragastric administration,the mice in each group were sacrificed,and their peripheral blood and bone marrow were collected.The peripheral blood white blood cell count,red blood cell count,platelet count and hemoglobin level were detected,the ROS levels of hematopoietic stem/progenitor cells in bone marrow were detected by flow cytometry,the colony forming unit-granulocyte and macrophage(CFU-GM)was detected by monolayer AGAR semi-solid culture method in vitro.Results Compared with the normal group,peripheral blood white blood cell count,red blood cell count,platelet count and hemoglobin level in the model group were significantly reduced(P<0.05),the levels of ROS in in bone marrow were significantly increased(P<0.05),and the number of CFU-GM in in bone marrow was significantly reduced(P<0.05).Compared with the model group,the peripheral blood leucocyte,erythrocyte,hemoglobin and platelet in low-dose and high-dose JBD groups were significantly increased(P<0.05),the level of ROS was significantly decreased(P<0.05),CFU-GM number in bone marrow were significantly increased(P<0.05),furthermore,the improvement of each index in the low-dose
