摘  要:目的探讨谷氨酸-脯氨酸-亮氨酸富集蛋白1(PELP1)在二乙基亚硝胺(DEN)诱导糖尿病小鼠肝癌形成中的表达及意义。方法6~8周龄雄性C57BL/6小鼠32只,随机分为4组:正常对照组(Control组),糖尿病组(DM组),二乙基亚硝胺组(DEN组),糖尿病联合二乙基亚硝胺组(DM+DEN组),每组8只。采用高脂饲料饲养构建糖尿病小鼠模型,并通过DEN诱导糖尿病小鼠肝癌形成。实验末,观察各组小鼠肝脏的大小、颜色、质地以及结节形成状况,记录肝脏质量、肝脏癌变只数和表面癌结节数,计算肝脏指数及癌变率;采用可见分光光度法检测各组小鼠血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)、甘油三酯(TG)和总胆固醇(TC)的含量;实时荧光定量PCR(qPCR)检测肝脏PELP1 mRNA转录水平。结果DEN组和DM+DEN组小鼠在造模12周后肝脏表面均出现结节。与Control组比较,DM组、DEN组和DM+DEN组小鼠肝脏指数和血清ALT和AST的含量明显升高(P<0.05);DM+DEN组与DM、DEN组比较,小鼠肝脏指数、肝脏癌变率、肝表面癌结节数和血清ALT、AST含量均显著升高(P<0.05);与Control组比较,DM+DEN组和DM组小鼠血清TG、TC含量明显升高(P<0.05)。与Control组比较,DM+DEN、DEN组小鼠肝脏组织中PELP1 mRNA表达显著升高(P<0.05),且DM+DEN组与DM、DEN组比较,差异有统计学意义(P<0.05)。结论糖尿病小鼠的肝脏功能出现损伤,DEN使糖尿病小鼠的肝功能进一步恶化,并诱发癌变,PELP1可作为肝癌发生的标志因子,在DEN诱导糖尿病小鼠肝癌形成中起重要作用。Objective To investigate the expression and significance of proline-,glutamic acid-,and leucine-rich protein 1(PELP1)in hepatic carcinoma induced by diethylnitrosamine(DEN)in diabetic mice.Methods Diabetic mice model was established by high fat diet,and hepatic carcinoma was induced by DEN in diabetic mice.32 male C57BL/6 mice aged 6-8 weeks were randomly divided into 4 groups:normal control group(Control group),diabetes mellitus group(DM group),diethylnitrosamine group(DEN group),and diabetes mellitus combined with diethylnitrosamine group(DM+DEN group),with 8 mice in each group.At the end of the experiment,the size,color,texture and nodular formation of the liver in each group were observed.The liver mass,number of liver cancerous lesions and number of surface cancer nodules were recorded.The liver index(liver mass/body mass×100%)and the carcinogenesis rate(number of cancerous lesions/total number×100%)were calculated.The concentrations of alanine aminotransferase(ALT),aspartate aminotransferase(AST),triglyceride(TG)and total cholesterol(TC)in serum of mice were determined by visible spectrophotometry.Real-time fluorescent quantitative PCR(qPCR)was used to detect the transcription level of PELP1 mRNA in the liver.Results Nodules appeared on the liver surface of mice in DEN group and DM+DEN group 12 weeks after modeling.Compared with control group,liver index and serum ALT and AST concentrations in DM group,DEN group and DM+DEN group were significantly increased(P<0.05).Compared with DM and DEN groups,liver index,liver carcinogenesis rate,number of liver surface cancer nodules and concentrations of ALT and AST in serum of mice in DM+DEN group were significantly increased(P<0.05).Compared with control group,the concentrations of TG and TC in serum of mice in DM+DEN group and DM group were significantly increased(P<0.05).Compared with control group,the expression of PELP1 mRNA in liver tissue of mice in DM+DEN group and DEN group was significantly increased(P<0.05),and the difference between DM+DEN group and DM
