摘  要:目的:检测转移相关蛋白1(MTA1)在甲状腺癌细胞系和甲状腺癌组织的表达,探讨其是否参与介导了甲状腺癌细胞的上皮-间充质转化(EMT)。方法:采用蛋白质印迹法(Western blot)检测MTA1在5个甲状腺癌细胞株和来自河南大学第一附属医院的153例甲状腺癌组织的蛋白质表达水平。应用小分子干扰RNA(siRNA)抑制MTA1,光学显微镜下观察对细胞形态的影响,Western blot检测对EMT信号通路蛋白的影响。采用T检验和方差分析进行统计学分析。结果:MTA1在未分化甲状腺癌细胞系HTh83和KMH-2中高表达,而在乳头状癌细胞系TPC-1、K1和滤泡状癌细胞系FTC133中低表达或者不表达。MTA1在30%的甲状腺癌组织中高表达。MTA1在颈淋巴结阳性者中的表达高于在颈部淋巴结阴性者中的表达(χ^(2)=44.449,P<0.01),差异有统计学意义,在有远处转移者中的表达高于在无远处转移者中的表达(χ^(2)=14.807,P<0.01),差异有统计学意义,与原发灶大小无明显相关(χ^(2)=0.627,P>0.05),差异无统计学意义。应用siRNA抑制MTA1在HTh83细胞中的表达,能够使该细胞系由间质样转变成上皮样生长。Western blot检测发现抑制MTA1的表达可以使N-Cadherin的表达明显升高,而同时E-Cadherin表达明显降低。结论:MTA1可能在未分化甲状腺癌细胞中表达水平更高,与转移扩散关系密切,可能参与了甲状腺癌的EMT。Objective To detect the expression of metastasis-associated protein 1(MTA1)in thyroid cancer cell lines and investigated the role of MTA1 in epithelial-mesenchymal transition(EMT)in thyroid cell lines.Methods Western blotting was used to examine the expression of MTA1 in 5 thyroid cancer cell lines,as well as in 153 cases of thyroid cancer tissue.The expression of MTA1 in thyroid cancer HTh83 cell line was knocked down by siRNA,and the effect of MTA1 silencing on cell morphology was observed under light microscopy and the effect on the EMT signaling proteins was investigated by Western blotting.Results MTA1 was detected in 4 out of 5 thyroid cancer cell lines.MTA1 was detected in 46 out of 153 thyroid cancer tissue.MTA1 was up-regulated in undifferentiated HTh83 and KMH-2 cell lines,and donw-regulated in papillary cancer TPC-1 and follicular FTC133 cell lines,and was undetectable in papillary cancer K1 cell line.HTh83 cell line displayed mesenchymal morphology in cell culture.However,knockdown of MTA1 in HTh83 cell line exhibited epithelial-specific morphology.Moreover,knockdown of MTA1 led to a clear increase of E-Cadherin and a decrease of N-Cadherin in the cells.Conclusion MTA1 may be up-regulated in undifferentiated thyroid cancer cells,and in advanced thyroid cancer.MTA1 may be involved in the EMT of thyroid cancer.
