摘 要:目的:观察T淋巴瘤侵袭转移诱导因子1(Tiam1)对鼻咽癌细胞增殖、侵袭和转移的影响。方法:选取2017年6月至2019年6月郑州大学第一附属医院咽喉头颈外科收集的102例鼻咽癌和癌旁组织作为研究对象,采用荧光定量聚合酶链反应(PCR)分析肿瘤组织和癌旁组织中Tiam1 mRNA表达。采用荧光定量PCR分析NP69、HK-1和CNE-2Z细胞系中Tiam1 mRNA表达水平。采用慢病毒介导Tiam1短发卡RNA(shRNA)和对照shRNA感染CNE-2Z鼻咽癌细胞,建立对照细胞系和Tiam1敲降细胞系,分别为对照组和Tiam1 KD组。采用噻唑蓝(MTT)法分析两组细胞增殖能力;采用Transwell分析两组细胞侵袭能力;采用异体移植瘤实验分析两组细胞体内生长和转移能力。组间计量资料比较采用t检验。结果:癌旁组织Tiam1 mRNA表达水平(1.36±0.29)明显低于鼻咽癌组织Tiam1 mRNA表达水平(3.98±0.41),差异有统计学意义(t=4.901,P<0.05)。正常鼻咽上皮细胞Tiam1 mRNA表达水平(1.20±0.26)明显低于鼻咽癌细胞系HK-1和CNE-2Z Tiam1 mRNA表达水平(2.98±0.31、3.16±0.29),差异有统计学意义(t=3.192,P<0.05;t=4.209,P<0.05)。对照组细胞吸光度(A)值(1.95±0.22)高于Tiam1 KD组细胞A值(1.37±0.19),差异有统计学意义(t=3.104,P<0.05)。对照组细胞体内生长45 d后肿瘤体积[(1832.37±281.24)mm 3]高于Tiam1 KD组细胞[(1209.31±145.19)mm 3],差异有统计学意义(t=4.016,P<0.05)。对照组细胞侵袭数量[(109.39±9.31)个]明显高于Tiam1 KD组细胞侵袭数量[(52.19±7.56)个],差异有统计学意义(t=5.109,P<0.05)。对照组细胞体内淋巴结转移数量[(12.43±3.27)个]明显高于Tiam1 KD组细胞[(6.19±2.01)个],差异有统计学意义(t=3.120,P<0.05)。结论:Tiam1在鼻咽癌组织中呈高表达,参与鼻咽癌细胞增殖、侵袭和转移。Objective To investigate the effect of T-lymphoma invasion and metastasis inducing factor-1(Tiam1)on proliferation,invasion and metastasis of nasopharyngeal carcinoma cells.Methods Totally,102 cases of nasopharyngeal carcinoma and adjacent tissues from June 2017 to June 2019 collected in our hospital were selected as the research objects.The expression of Tiam1 mRNA in tumor tissues and adjacent tissues were analyzed by fluorescence quantitative polymerase chain reaction(PCR).The expression levels of Tiam1 mRNA in NP69,HK-1 and CNE-2Z cell lines were analyzed by fluorescence quantitative PCR.The control cell line and Tiam1 knockdown cell line were established by infecting with lentivirus mediated Tiam1 short hairpin RNA(shRNA)and control shRNA as control group and Tiam1 KD group in CNE-2Z nasopharyngeal carcinoma cells,respectively.The proliferation ability of cells in the two groups was analyzed by methyl thiazolyl tetrazolium(MTT)assay and xenograft tumor model.The invasion ability of cells in the two groups was analyzed by Transwell.The in vivo metastasis ability of cells in the two groups was analyzed by xenograft tumor experiment.T test was used to compare the measurement data between the groups.Results The expression level of Tiam1 mRNA in adjacent tissues(1.36±0.29)was significantly lower than that in nasopharyngeal carcinoma tissues(3.98±0.41,t=4.901,P<0.05).The expression of Tiam1 mRNA in normal nasopharyngeal epithelial cells(1.20±0.26)was significantly lower than that in nasopharyngeal carcinoma cell lines HK-1 and CNE-2Z(2.98±0.31,3.16±0.29,t=3.192,P<0.05;t=4.209,P<0.05).The absorbance(A)value in control group(1.95±0.22)was higher than that in Tiam1 KD group(1.37±0.19,t=3.104,P<0.05).The tumor volume in control group was(1832.37±281.24)mm3,significantly higher than that in Tiam1 KD group[(1209.31±145.19)mm3,t=4.016,P<0.05].The number of invasive cells in control group[(109.39±9.31)cells]was significantly higher than that in Tiam1 KD group[(52.19±7.56),t=5.109,P<0.05].The number of lymph nod
