摘 要:目的分析枸杞多糖(LBP)对老年痴呆(AD)大鼠海马区核苷酸结合寡聚化结构域样受体3(NLRP3)、天冬氨酸特异性半胱氨酸蛋白酶-1(Caspase-1)和白介素-1β(IL-1β)含量的影响,探讨LBP延缓衰老的作用机制。方法制备AD大鼠模型,因干预措施不同,将大鼠安排为4组:对照组、AD组、LBP组及吡乙酰胺组,每组6只。LBP组及吡乙酰胺组的大鼠1天1次分别灌胃100 mg/kg LBP和400 mg/kg吡乙酰胺,正常组和AD组1天1次分别灌胃等质量体积的生理盐水,干预时间为4周。将大鼠投入水迷宫中检测其大脑学习、记忆功能的变化;Western blot分析大鼠海马区NLRP3、caspase-1及IL-1β水平的改变。结果相对于对照组,AD组大鼠的大脑学习、记忆功能减弱(P<0.05);海马区NLRP3(1.542±0.056)、Caspase-1(2.960±0.095)和IL-1β(3.026±0.074)水平的上升(均P<0.05)。相对于AD组,LBP组大脑学习、记忆功能增强(P<0.05);海马区NLRP3(0.872±0.034)、Caspase-1(1.878±0.079)及IL-1β(1.904±0.066)水平的下降(均P<0.05);LBP组及吡乙酰胺组比较差异无统计学意义(P>0.05)。结论 LBP能增强AD大鼠的学习和记忆功能,这与其减少AD大鼠海马区NLRP3、Caspase-1及IL-1β的水平相关。Objective To explore the effect of Lycium barbarum polysaccharides(LBP) on the contents of NOD-like receptor protein 3(NLRP3),Caspase-1 and interleukin-1β(IL-1β) in hippocampus of Alzheimer’s disease(AD) rats,study the mechanism of LBP in delaying aging. Methods AD rat model was established. Based on the different interventions,rats were divided into four groups:control group,AD group,LBP group and pyracetamide group. LBP group and pyracetamide group was respectively given100 mg/Kg LBP and 400 mg/Kg pyracetamide by gavage once a day. The normal group and AD group were only given the same volume of normal saline once a day. The intervention time was four weeks. The rats were put into water maze to detect the changes of learning and memory function,and the levels changes of NLRP3,caspase-1 and IL-1 β in hippocampus were analyzed by Western blot. Results Compared as the control group, the learning and memory functions of AD group were decreased(P<0.05),and the levels of NLRP3(1.542±0.056),Caspase-1(2.960±0.095)and IL-1β (3.026±0.074) increased in hippocampus(all P<0.05). Compared as AD group,the learning and memory functions of LBP group were enhanced(P<0.05),and the levels of NLRP3(0.872 ±0.034),Caspase-1(1.878±0.079) and IL-1β (1.904 ±0.066) in hippocampus were decreased in LBP group(all P<0.05). There was no statistically significant difference between LBP group and pyracetamide group(P>0.05). Conclusion LBP can enhance the learning and memory function of AD rats,which is related to the decrease of NLRP3,Caspase-1 and IL-1β levels in hippocampus of AD rats.
