摘 要:研究了常压室温等离子体(Atmospheric and Room Temperature Plasma,ARTP)诱变技术选育多拉菌素高产菌。建立了利用24孔板进行批量发酵培养和酶标仪快速检测筛选方法,获得了高产菌株的形态变化特性,筛选到了多拉菌素发酵高产菌S.avermitilis 3D5。在5 L罐上实验结果显示:菌株3D5在初级代谢期具有较高的细胞呼吸活力,能够快速生长,菌体最大比生长速率为0.36 h^(-1),相对于出发株提高了38.46%,底物糖转化率比出发菌株提高10.31%;多拉菌素高产菌3D5最终效价达531.93 mg/L,产量提高了36.34%,表明该高产菌株3D5具有较高工业化应用价值。This work focuses on the high through screening of doramectin high-yield strains with atmospheric and room temperature plasma(ARTP)mutagenesis technology.The optimized liquid microtiter plate culture system and the rapid detection method based on microplate reader were established for screening mutations of Streptomyces avermitilis,morphological characteristics of the strain with high yield were obtained,and the high-yield mutant strain S.avermitilis 3D5 was obtained.The verification results in the 5 L fermenter show that the mutant strain revealed higher respiratory metabolic activity and the maximum specific growth ratereached 0.36 h^(-1),which was 38.46%higher than that of the original strain.The yield of doramectin reached 531.93 mg/L,which was 36.34%higher than the original strain.At the same time,the conversion rate of substrate sugar to doramectin increased by 10.31%.The results showed the high-yield strain 3D5 had high industrial application value.
