摘 要:目的研究白花前胡乙素抑制卵巢癌细胞SK-OV-3增殖的作用及机制。方法通过CCK-8细胞活力检测法对不同浓度的白花前胡乙素作用24、48 h对卵巢癌细胞SK-OV-3增殖的影响;划痕实验考察20、40、60μmol/L白花前胡乙素对卵巢癌细胞SK-OV-3迁移的影响;20、40、60μmol/L白花前胡乙素给药作用24 h后,RT-qPCR检测c-Myc、细胞周期蛋白(Cyclin)D1等增殖基因mRNA的表达水平及胆固醇调节元件结合蛋白(SREBP)1c、脂肪酸合成酶(FASN)等肿瘤细胞能量代谢关键基因mRNA的表达水平;Western印迹检测SREBP1c、FASN等蛋白的表达水平。结果不同浓度的白花前胡乙素对卵巢癌细胞SK-OV-3增殖均具有抑制作用,且随着白花前胡乙素浓度升高,作用时间增长,对卵巢癌细胞SK-OV-3增殖的抑制作用增强;20、40、60μmol/L白花前胡乙素给药作用后能够抑制卵巢癌细胞SK-OV-3迁移,降低c-Myc、CyclinD1 mRNA基因表达效果,同时使其蛋白表达水平在一定程度上被降低。结论白花前胡乙素对卵巢癌细胞SK-OV-3增殖具有抑制作用,其作用机制可能是通过SREBP1c/FASN信号通路调节卵巢癌细胞SK-OV-3能量代谢从而抑制其增殖。Objective To study the inhibitory effect of praeruptorin B on SK-OV-3 proliferation in ovarian cancer cells and explore its mechanism.Methods CCK-8 cell viability assay was used to investigate the effects of different concentrations(10,20,40,60,80,100μmol/L)of praeruptorin B on SK-OV-3 proliferation in ovarian cancer cells for 24 and 48 h and the effect of 20,40,60μmol/L preflorin on SK-OV-3 migration in ovarian cancer cells was investigated by scratch test.After the administration of 20,40 and 60μmol/L praeruptorin B for 24 h,real-time fluorescence quantitative PCR(RT-qPCR)was used to detect the mRNA expressions of proliferating genes such as c-Myc and CyclinD1 and the mRNA expressions of key genes of energy metabolism such as SREBP-1c and FASN in tumor cells and Western blot was used to detect the expressions of SREBP-1c,FASN proteins.Results The proliferation of SK-OV-3 in ovarian cancer cells was inhibited by different concentrations of praeruptorin B,and the proliferation of SK-OV-3 in ovarian cancer cells was inhibited by increasing the concentration of praeruptorin B and the action time.20,40,60μmol/L praeruptorin B inhibited SK-OV-3 migration in ovarian cancer cells,and reduced the expressions of c-Myc,CyclinD1,SREBP-1c and FASN mRNA and the expressions of SREBP-1c,FASN proteins were reduced.Conclusions Praeruptorin B has an inhibitory effect on SK-OV-3 proliferation of ovarian cancer cells,and its mechanism might be that SREBP-1c/FASN signaling pathway regulates SK-OV-3 energy metabolism of ovarian cancer cells to inhibit its proliferation.
