补肾活血固齿方对MC3T3-E1细胞PDGF表达的影响

摘　　要：目的研究补肾活血固齿方对小鼠颅顶前成骨细胞(MC3T3-E1细胞)血小板生长因子(platelet derived growth factor,PDGF)表达的影响,探讨该方剂诱导成骨的作用机制。方法 MC3T3-E1细胞分别用10%补肾活血固齿方含药血清、无药血清、胎牛血清,培养24h、48h、72h,ELISA法检测细胞上清液中PDGF的表达。结果含药血清组培养上清液光密度值明显高于无药血清组和胎牛血清组,统计结果显示差异有显著性(P<0.05)。结论补肾活血固齿方能促进MC3T3-E1细胞分泌PDGF,参与诱导成骨作用。Objective To investigate the effect of Bushen Huoxue Guchi formula on the expression of platelet derived growth factor(PDGF) in MC3T3-E1 cells(pre-osteoblasts in mice calvarium). Methods MC3T3-E1 cells were cultured respectively by 10% herb-contained serum,blank-contained serum and fetal bovine serum for 24 h,48 h and 72 h. The expression of PDGF was detected in cell culture supernatant through ELISE assay. Results The optical density of the culture supernatant of the 10% herb-contained serum group was significantly higher than that of the blank-contained serum group and the fetal bovine serum group(P <0.05). Conclusion Bushen Huoxue Guchi formula can involve in inducing osteogenesis by upregulating the expression of PDGF in MC3T3-E1 cells.
