摘　　要：目的:探讨CLDN1在肝癌组织中的表达及其过表达对肝癌细胞恶性表型的影响。方法:通过免疫组化法及PCR法测定肝癌组织与癌旁组织中CLDN表达水平,建立CLDN1过表达细胞系,测定CLDN1过表达细胞株增殖能力、肿瘤细胞成球能力及干性相关标志物水平。结果:CLDN1在肝癌组织中表达阳性率为65.2%(58/89),在癌旁组织中表达阳性率为25.5%(14/55),差异具有统计学意义(P<0.001);CLDN1过表达肝癌细胞株增殖能力、肿瘤细胞成球能力均明显增强,并且肿瘤干性相关分子标志物水平明显提高。结论:CLDN1过表达可影响肝癌细胞干性获得,提示CLDN1表达水平与肝癌发生发展密切相关。Objective:To explore the expression of CLDN1 in hepatocellular carcinoma,and to explore the effect of CLDN1 overexpression on the malignant phenotype of liver cancer cells.Methods:The expression levels of CLDN in liver cancer tissues and adjacent tissues were determined by immunohistochemistry and PCR methods,and the CLDN1 overexpression cell line was established.The proliferation ability of CLDN1 overexpression cell lines,tumor cell sphere formation ability and stem-related markers were determined.Results:The positive rate of CLDN1 expression in liver cancer tissues was 65.2%(58/89),and the positive rate in adjacent tissues was 25.5%(14/55),the difference was statistically significant(P<0.001).CLDN1 overexpression of hepatocellular carcinoma cell line proliferative ability,tumor cell sphere formation ability were significantly enhanced,and tumor stem related molecular markers were significantly increased.Conclusion:The overexpression of CLDN1 can affect the dryness of liver cancer cells,suggesting that the expression level of CLDN1 is closely related to the occurrence and development of liver cancer.
